猕猴桃雌雄株分子标记的收集、评价与应用

Collection, Evaluation and Application of Molecular Markers for Kiwifruit Sex Identification

摘要: 【目的】猕猴桃为雌雄异株多年生藤本果树,早期的性别鉴定有利于加速选育进程,节约育种成本。【方法】收集了前人已开发的3组猕猴桃性别鉴定标记,SCAR标记SmX和SmY1,SNP标记L11和L51以及RAPD标记S1032850,并在41份已知性别的猕猴桃材料上进行通用性验证。【结果】SmX、L11、L51和S1032850标记未表现出性别特异性,而SmY1标记能准确鉴别猕猴桃性别,雄株鉴定准确率为85%,雌株鉴定准确率为100%,且在实生幼苗能扩增出稳定条带,操作简便。【结论】SmY1标记高效准确,可作为猕猴桃性别鉴定的主要分子标记广泛应用。

Abstract: 【Objective】 Kiwifruit is a dioecious perennial vine fruit tree, and early sexual identification is beneficial to accelerate the breeding process, and saves breeding costs. 【Method】 Three sets of kiwifruit sex identification markers, SCAR markers SmX and SmY1, SNP markers L11 and L51 and RAPD marker S1032850, which had been developed previously, were used for universal verification on 41 kiwifruit materials of known sex. 【Result】 The SmX, L11, L51 and S1032850 markers did not show gender specificity, while the SmY1 marker could accurately identify the sex of kiwifruit. The accuracy rate of male plant identification was 85%, and the accuracy rate of female plant identification was 100%, and it can amplify stable bands in young seedlings, easy to operate. 【Conclusion】 SmY1 marker is efficient and accurate, and can be widely used as the main molecular marker for gender identification of kiwifruit.
