β-胡萝卜素对肉用母牛卵泡发育和繁殖轴相关基因表达的影响

Effects of β-Carotene Supplementation on Follicular Development and Reproductive Axis Related Gene Expression in Beef Heifer

摘要: 【目的】旨在考察饲粮中添加β-胡萝卜素对肉牛后备母牛卵泡发育的影响。【方法】选取12头体重相近(285 kg±12.3 kg)15月龄健康母牛(西杂牛♂×犏牛♀),随机分为2组,每组6头。对照组饲喂基础饲粮,处理组在对照组的基础上额外添加β-胡萝卜素1 200 mg/头/天。正式试验3个月。最后一天里每隔15 min采集血清样品对黄体生成素(LH)和卵泡生成素(FSH)进行测定,用超声仪对卵泡进行检测,后将所有牛只屠宰,采集垂体、下丘脑以及卵巢对繁殖轴相关基因表达进行测定。【结果】添加β-胡萝卜素可提高血清中LH含量(P<0.001);提高了下丘脑中Kiss1GPR54GnRHEGR1CaMKII以及ERK1/2等基因表达(P<0.05);提高了垂体中LHβ的表达(P<0.05);提高了卵巢中ERα表达,提高了大、中卵泡的比例(P<0.001)。【结论】β-胡萝卜素通过提高Kiss1/GPR54表达量激活ERK1/2信号途径,提高垂体中LHβ表达,促进LH分泌,促进卵泡发育。

Abstract: 【Objective】 The purpose of this experiment was to investigate the effect of dietary β-carotene supplementation on follicular development in beef heifer.【Method】 Twelve healthy 15-month-old heifers(Simmental crossbred♂×yak crossbred♀) of similar weight (285 kg±12.3 kg) were selected and randomly divided into 2 groups,and each treatment had 6 heads.The control group was fed basal diet,and each cattle in the treatment group was supplemented with an additional β-carotene 1 200 mg/d on the basis of the control group.Formal experiment lasted 3 months.On the last day of the formal experiment,eight serum samples in a 15 min interval were collected to measure luteinizing hormone (l h) and follicle stim-ulating hormone (fsh),ultrasound equipment was used to detect the status of follicles and then all six animals were slaughtered,genes related to the reproductive axis in the pituitary,hypothalamus,and ovary were determined.【Result】 The results showed that the addition of β-carotene increased the lh content in serum (P<0.001).the addition of β-carotene increased the expression of Kiss1, GPR54, GNRH, EGR1, CAMKII,and ERK1/2 in the hypothalamus (P<0.05).The addition of β-carotene increased the expression of LHβ in the pituitary (P<0.001).The addition of β-carotene increased ERα expression in the ovary (P<0.001) and increased the proportion of large and medium follicle (P<0.001).【Conclusion】 β-carotene supplementation activates Kiss1/GPR54 increases the expression of LHβ in the pituitary,promotes LH secretion,through ERK1/2 signal and finally promotes follicular development.
