基于16SrRNA基因测序技术研究低聚异麦芽糖对奶牛瘤胃细菌菌群的影响

Effect of Isomaltooligosaccharides on Rumen Racterial Flora in Dairy Cows Based on 16SrRNA Gene Sequencing Technology

摘要: 【目的】本试验旨在利用16SrRNA基因测序技术研究低聚异麦芽糖对奶牛瘤胃菌群的影响。【方法】试验采用两阶段交叉设计方法,将4头泌乳中国荷斯坦泌乳奶牛随机分为两组,第一阶试验结束,经过恢复期后,再将试验组和对照组交叉,由之前对照组变为试验组。试验组添加60 g/(d·头)的低聚异麦芽糖。【结果】结果表明日粮添加低聚异麦芽糖后,奶牛瘤胃细菌菌群多样性无显著影响。门水平上分析,疣微菌门丰度显著增加(P<0.05)。属水平上分析,饲喂低聚异麦芽糖,奶牛瘤胃液Comamonadaceae_NA(未注释丛毛单胞菌属)丰度极显著提高(P<0.01),WCHB1-25_NA丰度极显著增加(P<0.01),Ruminobacte(瘤胃杆菌属)丰度显著增加(P<0.05),Salinibacterium(盐细菌属)丰度显著增加(P<0.05),Lactobacillus(乳酸杆菌属)丰度显著降低(P<0.05)。Acidaminococcus(氨基酸球菌属)、Aeromicrobium(气微菌属)、ActinotaleaAdlercreutzia菌属丰度有增加趋势(0.05<P<0.1),TMEG_NA菌属丰度有降低的趋势(0.05<P<0.1)。【结论】在本试验条件下,日粮添加低聚异麦芽糖对奶牛瘤胃菌群多样性无显著影响,纤维分解菌丰度影响差异不显著,淀粉分解菌丰度增加、乳酸生成菌丰度降低。

Abstract: 【Objective】 This experiment aims to study the effect of isomaltooligosaccharides on rumen flora in dairy cows by using16SrRNA gene sequencing technology. 【Method】 Four lactating Chinese Holstein dairy cows were randomly divided into two groups by two-stage crossover design method. After the first-stage experiment, the test group and the control group were crossed and changed from the former control group to the test group. The test group was supplemented with 60 g/(d-head) isomaltooligosaccharides in diet. 【Result】 The results showed that flora diversity of rumen bacteria in dairy cows was not different after adding isomaltooligosaccharides in diet. At the phylum level, the richness of Verrucomicrobiaincr-eased significantly(P<0.05). At the level of the genus, The abundance of Comamonadaceae_NA in the rumen fluid increased significantly (P<0.01), WCHB1-25_NA abundance increased significantly (P<0.01),Ruminobacte abundance increased significantly (P<0.05), Salinibacterium abundance increased signifi-cantly (P<0.05) and Lactobacillus abundances significantly decreased (P<0.05). The abundance of Aci-daminococcus,Aeromicrobium,Actinotalea and Adlercreutzia has an increasing trend (0.05<P<0.1),and the abundance of TMEG_NA genera has a tendency to decrease (0.05<P<0.1). 【Conclusion】 In this exp-eriment,flora diversity of rumen bacteria in dairy cows was not different after adding isomaltooligosaccha-rides in diet, and there was no significant difference in the abundance of cellulolytic bacteria in the rumen of dairy cows, the abundance of starch decomposing bacteria and the decrease of the abundance of lactic acid producing bacteria.
