Pyrrosia petiolosa对小鼠皮肤伤口愈合作用研究

Effect of Pyrrosia petiolosa Extract on Dermal Wound Healing in Mice

摘要: 【目的】评估有柄石韦(Pyrrosia petiolosaChristChing, P. petiolosa)水提取物(Pyrrosia petiolosa aqueous extract,PPAE)与乙酸乙酯提取物(Pyrrosia petiolosa ethyl acetate extract,PPEAE)对小鼠皮肤伤口愈合的影响。【方法】构建小鼠皮肤切除创伤模型并随机分为4个处理组(阴性对照组、云南白药组、PPAE组、PPEAE组),处理14 d后检测小鼠伤口愈合率、羟脯氨酸含量,对伤口组织进行HE染色观察成纤维细胞上皮形成程度,并检测PPAE对供试病原菌的抑制效果。【结果】PPAE组小鼠创面伤口愈合率显著高于对照组(80.3% vs 45.6%)(P<0.05)。PPAE处理后创面组织总羟脯氨酸含量增至4.1 μg/mg,且PPAE给药组小鼠伤口组织的组学分析也显示其上皮化程度增加,成纤维细胞增殖增强,同时PPAE对供试菌株表现出抗菌活性。PPEAE组伤口愈合率、羟脯氨酸含量和病理组织切片结果与对照组差异不显著。【结论】P. petiolosa 提取物PPAE的局部外用可促进小鼠伤口愈合。

Abstract: 【Objective】 To evaluate the effects of Pyrrosia petiolosa aqueous extract (PPAE) and Pyrrosia petiolosa ethyl acetate extract (PPEAE) on dermal wound healing in mice. 【Method】 A mouse skin resection wound model was constructed and randomly divided into four groups (negative control group, Yunnan White drug group, PPAE group, PPEAE group). After the treatment for 14 days, the wound healing rate and hydroxyproline content of the mice were detected. HE staining was used to observe the degree of fibroblast epithelial formation, and the inhibitory effect of PPAE on the tested pathogens was tested.【Result】 The wound healing rate of mice in the PPAE group was significantly higher than that in the control group (80.3% vs 45.6%) (P<0.05). After PPAE treatment, the total hydroxyproline content of the wound tissue increased to 4.1 μg/mg, and the histological analysis of the wound tissue of the PPAE group also showed an increase of epithelialization and the proliferation of fibroblasts. Meanwhile, PPAE showed antibacterial activity against the tested strains. The wound healing rate, hydroxyproline content and pathological tissue sections of the PPEAE group were not significantly different from the control group. 【Conclusion】 The topical application of P.petiolosa extract PPAE can promote wound healing in mice.
