The Specific Molecular Marker for Identification of Plasmodiophora brassicae Pathotype 4

摘要: 【目的】根肿病已成为我国油菜等十字花科作物的主要病害。芸薹根肿菌(Plasmodiophora brassicae)作为专性寄生菌,单孢分离和培养技术要求较高,制约着根肿菌生理小种的分子鉴定,建立一种快速、方便、准确的检测方法,将为田间根肿菌生理小种的监测、预警及综合防控等提供科学依据。【方法】采用转录组测序,筛选根肿菌侵染感病白菜(Brassica rapa)下4号生理小种区别于其他供试小种的差异表达基因,通过常规PCR进行验证。【结果】挖掘获得存在于根肿菌4号生理小种,而供试2、5、7、9、10和11号小种中不存在的2个分子标记基因PBRA_000030和Novel00510,通过降低PCR扩增循环数为25这一优化方式,达到特异鉴定肿根中4号优势生理小种的目的。【结论】针对我国根肿菌优势生理小种4号,建立了一种操作简便、特异性强、适用范围广的方法。

Abstract: 【Objective】 In recent years, the clubroot has become a major disease of Brassica napus and other cruciferous crops in China. Plasmodiophora brassicae, as an obligate parasitic pathogen, is difficult for the single spore isolation and culture technology, which restricts the molecular identification of pathotype. The detection system established in the study is simple, specific and widely applicable, which can provide reference for the monitoring, early warning and the comprehensive prevention and control of pathotype 4 in feild. 【Method】 Transcriptome sequencing was used to screen the differentially expressed genes of P. brassicae pathotyoe 4 from other pathotypes infecting Brassica rapa. These genes were verified by conventional PCR.【Result】The two molecular marker genes, PBRA_000030 and Novel00510 were found in pathotype 4, which different from other test pathotypes 2,5,7,9,10 and 11. The dominant physiological pathotype 4 in clubtoot could be identified by using PCR with about 25 amplification cycles.【Conclusion】A simple, specific and widely applicable method was established,basing on the molecular marker genes obtained from the dominant pathotype 4 in China.
