红螯螯虾转录组高通量测序及分析

Sequencing and Analysis of the Transcriptome of Cherax quadricarinatus

摘要: 【目的】为发掘和研究红螯螯虾功能基因SSR标记奠定基础。【方法】对红螯螯虾肝脏、精巢以及卵巢转录组测序选用新一代高通量测序技术Illumina Hi Seq 2000,同时借助生物信息学法展开基因表达谱探究及功能基因预测。【结果】肝脏组织获得47 629 414条clean reads,精巢组织获得47 018 968条clean reads,卵巢组织获得53 267 362条clean reads。所有reads组装后获得了67 369个Unigene。通过 GO分类, 16 989个 Unigene被分为3个种类,即生物学过程、细胞组分、分子功能。通过COG分类,4 697个 Unigene被分为25个种类。通过 KEGG分类,9 842个Unigene分属331个代谢途径。【结论】通过对红螯鳌虾肝脏等3个组织转录组的测序,获得了其相关基因的表达图谱及其功能分类。

Abstract: 【Objective】 To excavate and study the functional genes and SSR markers of red crayfish.【Method】 A new generation of high-throughput sequencing technology, Illumina Hi Seq 2000, was used to sequence the liver, testis, and ovary transcriptomes of Cherax quadricarinatus, and gene expression profiling and functional gene prediction were performed using bioinformatics methods. 【Result】 47 629 414 clean reads were obtained in the liver, 47 018 968 clean reads were obtained in the testes,and 53 267 362 clean reads were obtained in the ovarian tissues. After all reads were assembled, 67 369 Unigenes were obtained. Classified by GO, 16 989 Unigenes are divided into three categories, namely biological processes, cellular components, and molecular functions. By COG classification, 4 697 Unigenes are divided into 25 categories. By KEGG classification, 9 842 Unigenes belong to 331 metabolic pathways. This study lays a certain theoretical foundation for the better development and utilization of the resources of the red chelonian crayfish. 【Conclusion】 Gene expression profiling and its functional genes of C. quadricarinatus were obtained by using high-throughput sequencing technology.
