阿斯匹林在黄牛体内的药代动力学研究

STUDY ON PHARMACOKINETICS OF ASPIRIN IN BOS. TAURUS

摘要: 本文报道用高压液相色谱法研究了黄牛一次口服阿斯匹林片剂(100mg/kg)后药代动力学规律。测定条件:Nucleosil-C18,7μm,4×250mm不锈钢柱;流动相:乙腈-0.05%磷酸(30:70,V:V);紫外检测波长:237nm。主要动力学参数:t1/2Ke,4.295±1.043hr;Tmax,4.375±0.773hr;Cnax29.18±5.121μg/ml;AUC,366.532±86.554μg/ml.hr。

Abstract: This paper reports a method for determination of aspirin and its main metabolites--Salicylic acid in plasma of seven healthy bos. Taurus given aspirin as tablet, at a dose of 100 mgkg-1 body weight by HPLC. The detenmination factors are asfollows. Nucleosil-C18 as solid phase and the mobile phase consisted of acetonitrile-0.05% phosphate acid (30: 70, v: v) at a flow rate 1.0 mlmin-1. The wavelength of uvdector was 237nm. The experimental data were processed by using "WTS. TM. BAS" pharmacokinetics auto-program. The result indicates that the C-T curve is in accordance with one-compartment open model with first order absorption. The major parameters are:K=0.170±0.038 (hr-1) Ka=0.316±0.044 (hr-1) T1/2ke=4.295±1.043(hr) T1/2ka=2.1046±0.6458(hr) Tmax=4.375±0.773(hr) Cmax=29.181±5.121 (μgml-1) AUC=366.532±86.554 (μgml-1hr-1).
