利用AFLP标记鉴定小豆栽培型种质遗传多样性

Genetic Diversity in Cultivated Azuki Bean (Vigna angularis var.angularis) Germplasm Resources Revealed by AFLP Markers

摘要: 利用11对AFLP引物,对94份来自我国的小豆栽培型种质基因组DNA进行扩增,得到689条清晰的谱带,其中324(47.0%)条呈多态性,平均每对AFLP引物得到29.5条多态性带;平均遗传距离0.285,变异幅度为0.009~0.836。利用AFLP多态性数据进行NeiandLi遗传相似系数聚类分析,可将94份种质相互区分开,并划分为4个明显不同的组群,表明我国栽培型小豆种质存在较丰富的遗传多样性。4个组群的遗传多样性表现出明显的生育特性和生长习性趋同性,也表现出一定的地域相关性。

Abstract: 94 cultivated azuki bean (Vigna angularis var.angularis) germplasm from Chinese geographical regions are used for AFLP analysis with 11 informative primer pairs, and 689 unambiguous bands are created, 324 of which are polymorphic. One pair of AFLP primers has detected a mean of 29.5 polymorphic bands. All the 324 polymorphic bands are used to assess genetic distance among 94 azuki bean accession to form the genetic distance matrix by using Nei and Li's method. The mean genetic distance among materials is 0.285 with a range of 0.009 to 0.836 according to the matrix. Four clusters are detected by using UPGMA cluster analysis, based on the genetic distance matrix. According to the dentrogram formed by cluster analysis, 94 materials are distinctly clarified, revealing a enough genetic diversity for identification and classification of accessions within Vigna angularis var.angularis. Four clusters are clearly corresponded to the growth habit and the geographic origin of accessions.
