对‘早霞露’桃迟熟型芽变的研究

Study on the Late-ripening from ‘Zao Xia Lu’ Peach

摘要: 对特早熟桃‘早霞露’的迟熟型芽变进行了果实品质、过氧化物酶和酯酶同工酶、RAPD等方面的分析,结果表明该芽变为优变。芽变与‘早霞露’的酯酶同工酶存在显著差异,除酶活性不同外,与‘早霞露’相比,芽变缺少Rf0.43酶带,多一条Rf0.26酶带;而芽变与‘早霞露’过氧化物酶(POD)同工酶图谱相似。RAPD分析结果显示,在所用大量单引物和混合双引物的扩增片段中,未出现两者间片段有无的差异,仅有一条单引物S465的一个大分子量片段在芽变中扩增量一直多于对照。

Abstract: ‘Zao Xia Lu’ Peach and it's Late-ripening sport were studied on fruit quality, POD and EST isoenzyme electrophoresis and RAPD. The result shows that the sport is an excellent material. Even though their POD isoenzymes are much alike, there are significant differences in EST isoenzymes between ‘Zao Xia Lu’ and it's sport: on the whole, isoenzymes in the sport have stronger activity;moreover, compared with ‘Zao Xia Lu’, the band Rf 0.43 is absent while a new one Rf 0.26 appears in the sport. As for RAPD analysis, more than 300 arbitrary primers and 108 mixed primers are used to amplify genomes from ‘Zao Xia Lu’ and it's sport. As a result, the profiles amplified by RAPD in both materials are almost identical except the intensity difference of one fragment amplified by primer S465.
