外源DNA经减压渗透法处理水稻的变异后代同工酸分析

摘要: 以小麦、玉米、小米、高粱、狼尾草五个不同属的材料为外源DNA供体,经减压渗透转化受体紫稻。从获得的变异材料中各选一份在苗期、分蘖期、孕穗期、抽穗期四个时期用聚丙烯酰胺凝胶电泳(PAGE)和等电点聚焦电泳(PAG-IEF)分。析了过氧化物酶同工酶,结果发现各材料在所检测的四个时期均与受体紫稻存在不同程度的差异。这些差异多表现在变异后代材料在弱带和痕迹带的缺少和增加上。并且在变异材料与受体之间差异存在多种表现形式,在生理生化水平上表明了这些变异后代材料与受体紫稻间存在差异。

Abstract: By using polyacrylamide gel electrophoresis(PAGE) and polyacryla-mide gel isoelectric focusing(PAG-IEF). analysis on isoperoxidase was carried out for variant desendants obtained from transmitting isolated DNA of Triticum aestivum L., Zea may L., Setaria italica(L.) Beau V., Serghum vulgare Pers., Pennistum sp. into purple rice by decompress osmosis, a new plant molecular breed-ing technique. in four different growth stage, seedling, tillering, panicie formation and-heading. The results showed that isoperodases patterns were differert in a few weak and trace bands between purple rice and all variant descendants in the tested grawth stage, which indicated that the variants possess genetic base different that of the purple rice. In the end. the authors suggest that IEF be a powerful method for identifing genetic differences in plant.
