麦类作物染色体Giemsa C-带技术中的关键处理因子对带纹影响的研究

Influence of Bands on Critical Treatment Factors in the Giemsa-C Banding Procedure for Triticeae Chromosomes

摘要: 利用单因素和五因素二次回归正交旋转组合设计试验对影响麦类作物染色体C-带的5个因素(无水乙醇、0.2mol/LHCl、Ba(OH)2和2×SSC等的处理时间及Ba(OH)2冲洗水温)进行研究,结果表明:0.2mol/LHCl、Ba(OH)2和2×SSC等的处理时间对实验结果有极显著的影响,Ba(OH)2冲洗水温对实验结果的影响可以忽略不计。无水乙醇处理的作用是增加染色体对Ba(OH)2处理的耐受性。因此尽管在适当缩短Ba(OH)2处理时间的情况下,可省去无水乙醇处理这一过程,但麦类作物染色体C-带染色的较佳工艺为:无水乙醇处理12h,0.2mol/LHCl 60℃处理2.33~2.48min、Ba(OH)2 18℃处理7.11~7.39min,冲洗Ba(OH)2的水温控制在30~40℃,2×SSC的处理条件为56~60℃ 1.30~1.43h。

Abstract: The analyses were carried out by doubling orthogonal and rotate combination design for the five treated factors:0.2mol/L HCl, saturated Ba(OH)2 solution, temperature of water to wash Ba(OH)2 solution, and 2×SSC solution, which involved in the Giemsa-C banding procedure on the study Triticeae chromosomes. The statistical results indicate the most important factors to influence the banding pattern on the Triticeae chromosomes are the treatment of both the 0.2mol/L HCl and saturated Ba(OH)2 solution in the Giemsa-C banding technique. The function of alcohol treatment might be increasing the crop chromosome tolerance of saturated Ba(OH)2 treatment. Therefore, it is no necessity for the treatment of alcohol when a little treated time of saturated Ba(OH)2 solution is reduced, but in order to obtain better results in the Triticeae chromosome Giemsa C-banding procedure, it appears to be the adopting steps that are the treatments of 0.2mol/L HCl with range of 2.33 to 2.48 minutes in 60℃, and saturated Ba(OH)2 with 7.11 to 7.39 minutes in 18℃, and 2×SSC with 1.30 to 1.43 hour in 56~60℃. However, the alcohol treatment dodes not seem to be important in the C-banding technique while the treating time of Ba(OH)2 is reduced a little. The water temperature to wash Ba(OH)2 solution is better to control in 30~40℃ in order not to affect the final results.
