四川粗壮女贞苦丁茶中黄酮的提取、分离与初步鉴定

Extraction, Separation and Primary Identification of Flavonoids from Ligustrum robustum in Sichuan

摘要: 以粗壮女贞[Ligustrum robustum(Rxob.)BL.]苦丁茶为材料,选用乙醇浸提L9(34)正交实验,确定了从四川苦丁茶中提取黄酮的最佳工艺条件:乙醇浓度为80%,提取温度为80℃,乙醇用量为10倍于苦丁茶质量,浸提时间为4h,总黄酮的提取效果最好,物质的浸出率可达84.32%,所得干物质中黄酮总量为47.6803mg/g。对提取物进行分离和纯化,得到6种化合物,经薄层层析(GF254)和显色反应等初步定性鉴定,结果表明:化合物A为异黄酮;化合物B、D、F均为黄酮(醇);化合物C为二氢黄酮(醇);化合物E为花色甙。

Abstract: By using Ligustrum robustum(Rxob.) BL. in Sichuan as materials and the experiment of L9(34) by ethanol as extracting way, the optimum conditions of extracting flavonoids from Ligustrum robustum are obtained. When the condition is 80% of density of ethanol, 80℃ of extracting temperature, 10:1 ethanol volume and sample weight, 4h of extracting time, the extracting effect is better, the extracting rate can reach 84.32% and the total content of flavonoids is 47.6803 mg/g. Then the extraction are separated and purified to gain six kinds of compounds. Primary identification of the structure by thin-layer chromatographic procedure of silica GF254 and the color reactions show that compound A are isoflavones, compound B, D, F are all flavones(or flavonols), compound C are flavanones(or flavanonol), compound E are anthocyanins.
