绿粘帚霉原生质体制备和再生条件的研究

Production and Regeneration of Gliocladium virens (F051) Protoplast

摘要: 采取单因素实验对绿粘帚霉(Gliocladium virens F051)的原生质体制备和再生条件进行了研究,结果表明,绿粘帚霉(F051)原生质体制备和再生的最佳条件是:用培养基A培养F051分生孢子24h,MgSO40.6mol/L (再生用蔗糖0.6mol/L),磷酸氢二钠-柠檬酸缓冲液为缓冲系统,纤维素酶:蜗牛酶:溶菌酶=4mg/mL:2mg/mL:2mg/mL,在32℃水浴中酶解2h。

Abstract: The conditions of protoplast preparation and regeneration were studied on Gliocladium virens (F051)by means of one way experiment.The optimal conditions of protoplast preparation and regeneration of Gliocladium virens (F051)were showed as follows:the optimum conditions of F051 protoplast isolation and the regeneration are:conidiophores cultured for 24 h by using culture medium A,0.6 mol/L MgSO4 (regeneration used 0.6 mol/L sucrose)as osmotic stabilizer,the sodium hydrogen phosphate citric acid as the buffer system,the cellulase:Snailase:Lysozyme=4 mg/mL:2 mg/mL:2 mg/mL,in 32℃ water baths dige-sting 2 h.
