利用SRAP和SSR标记对汉中地区主要油菜品种的遗传多样性分析

Analysis of Genetic Diversity of Brassica napus Mainly Popularized in Hanzhong Based on SRAP and SSR Markers

摘要: [目的] 掌握陕西省汉中地区主要推广油菜品种的遗传多样性,了解其遗传背景,明确各品种间的亲缘关系,为油菜育种提供理论依据。[方法] 利用24对有多态性的SRAP标记和17对SSR标记,PCR扩增采用复性变温法,对34份陕西省汉中地区主要推广的油菜品种资源进行遗传多样性分析。[结果] 2种不同的复性温度都取得了好的扩增条带,SRAP标记共检测到145个等位变异,平均每个标记检测到6个等位变异,每个SRAP位点的遗传多态性信息含量在0.19~0.74之间,平均值为0.48。供试材料的遗传相似系数集中在0.56~0.94之间。SSR标记共检测到99个等位变异,平均每个标记检测到6个等位变异,每个SSR位点的遗传多态性信息含量在0.12~0.74之间,平均值为0.40,遗传相似系数集中在0.62~0.91之间。[结论] SRAP标记的遗传多态性比SSR标记高,SRAP标记聚类更接近系谱分析。陕西省汉中地区主要推广的油菜品种遗传相似度较高,亲缘关系较近。

Abstract: [Objective] This study was to understand the genetic diversity,genetic background and relationship of all rapeseed mainly popularized in Hanzhong,and to provide the theoretical basis for rapeseed breeding.[Method] We investigated genetic diversity of 34 main rapeseed strains mainly popularized in Hanzhong by utilizing 24pairs of SRAP makers and 17pairs of SSR.[Results] Total of 145alleles were detected by using SRAP markers and 99alleles were detected by using SSR.An average of 6alleles were obtained per pair of molecular markers.The PIC for SRAPs were between 0.19and 0.74with an average of 0.48,while the PIC for SSRs were between 0.12and 0.74with the average of 0.40.Cluster analysis showed that genetic similarity coefficient of 34testing materials of rapeseed centralized in the range of 0.56to 0.94.The genetic distances were between 0.62and 0.91.[Conclusion] The SRAP possessed higher potential in determining genetic diversity when compared with SSR.The cluster analysis results obtained by SRAP were closer to pedigree method.Overall,the tested rapeseed varieties have a close genetic relationship.
