云南地方猪种mtDNA D-loop区遗传多样性研究

The Genetic Diversity of Mitochondrial DNA D-loop in Yunnan Native Pigs

摘要: [目的] 为揭示云南7个地方猪种遗传多样性及其分布规律。[方法] 以云南7个地方猪品种(群体)为对象,研究了线粒体DNA D环高变区(mtDNA D-loop HVI)的序列多态性。[结果] 研究表明,在全长435bp的核苷酸序列中共检测到23个多态位点,由此界定了30个单倍型。云南地方猪种整体的单倍型多样度(Hd)和核苷酸多样度(Pi)分别为0.820和0.00451。在7个品种(群体)中,保山猪单倍型多样度最高(0.828),高黎贡山猪最低(0.644);滇南小耳猪的核苷酸多样度最高(0.00521),高黎贡山猪同样最低(0.00327)。由遗传变异贡献率和遗传独特性贡献所决定的总体遗传贡献率中,迪庆藏猪总体遗传贡献率最高(10.3429),高黎贡山猪和大河猪最低(均为1.0594)。[结论] 就总体单倍型丰富度效应而言,迪庆藏猪、滇南小耳猪、保山猪和撒坝猪对维持云南地方猪种遗传多样性呈正效应,而明光猪、高黎贡山猪和大河猪呈负效应。

Abstract: [Objective] To reveal the genetic diversity and distribution in seven native pig breeds/populations in Yunnan province. [Method] The mitochondrial DNA D-loop hypervariable region(mtDNA D-loop HVI) of Yunnan native pigs were investigated in this study. [Results] The results showed that 23 polymorphism sites were found in the full-length 435 bp which defined 30 haplotypes. The haplotype diversity(Hd) and nucleotide diversity(Pi) of Yunnan seven native pigs were 0.820 and 0.004 51 respectively. Among the seven native pig breeds/populations, the highest value of Hd was found in Baoshan pig(0.828) and the lowest in Gaoligongshan pig(0.644), the highest value of Pi(0.005 21) was found in Diannanxiaoer pig and the lowest Pi(0.003 27) was also found in Gaoligongshan pig. The total genetic rate unique determined by genetic variation and genetic unique that Diqing Tibetan pig(10.342 9) gave the highest contribution to the total genetic rate among Yunnan native pigs, the total genetic rate unique of both Gaoligongshan pig and Dahe pig were 1.059 4which provided the lowest contribution to Yunnan native pigs. [Conclusion] On the whole haplotype effects in terms of rich that Diqing Tibetan pig, Diannanxiaoer pig, Baoshan pig and Saba pig have the positive effect on the genetic diversity of Yunnan seven native pigs, Mingguang pig, Gaoligongshan pig and Dahe pig have the negative effect.
