黄花菜多糖的提取工艺及抗肿瘤活性研究

Study on the Optimal Extraction Technology and Anti-tumor Effect of Polysaccharide in Hemerocallis fulva

摘要: [目的] 研究黄花菜多糖的最佳提取工艺及黄花菜多糖抗肿瘤的活性。[方法] 采用苯酚-硫酸法测定黄花菜多糖的含量,在单因素试验的基础上,采用L9(34)正交试验,以黄花菜多糖提取率为指标,探讨了提取次数、提取温度、提取时间、料液比对黄花菜多糖提取率的影响,得到最佳工艺;采用体内肿瘤动物模型,在昆明种小鼠右腋皮下接种S180肿瘤细胞,灌胃后观察移植瘤抑制率,采用ABC-ELISA法检测S180荷瘤小鼠血清中IL-2、TNF-α水平。[结果] 黄花菜多糖的最佳工艺条件为:料液比1:25(g:mL),提取温度80℃,提取时间2 h,提取次数3次,黄花菜多糖的平均提取率为17.47%。黄花菜多糖可抑制小鼠S180移植瘤的生长,高剂量组(1.60 g/kg)抑瘤率为38.54%;黄花菜多糖能提高荷瘤小鼠血清中IL-2和TNF-α含量。[结论] 黄花菜多糖具有较明显的体内抗肿瘤活性,其机制可能与多糖能够提高机体的免疫功能有关。

Abstract: [Objective] This experiment was to study the optimal extraction technology and anti-tumor effect of polysaccharide in Hemerocallis fulva. [Methods] Determination of the content of polysaccharide in Hemerocallis fulva by phenol-sulfuric acid method. On the basis of single factor experiment, L9(34) orthogonal design was adopted to study the effects of extraction times, extraction temperature, extraction time and solid-liquid ratio on yield of polysaccharide. Using S180 tumor-bearing mice as model we observed mice transplanted tumor inhibition rate. The levels of IL-2 and TNF-α in serum were detected by antibody sandwich enzyme-linked inununosorbent assay(ELISA). [Results] The results showed that the optimal extraction process parameters were as followed:ratio of solid-liquid 1:25(g:mL), ex-traction temperature 80℃, extraction time 2 h, extracting 3 times. Under such conditions, the average extraction rate of polysaccharide in Hemerocallis fulva was 17.47%. Polysaccharides in Hemerocallis fulva had inhibited the growth of S180 transplanted tumor. The rate of inhibition was 38.54% in high-er-doses groups(1.60 g/kg). Polysaccharides in Hemerocallis fulva can improve contents of IL-2 and TNF-α in serum of mice with tumors. [Conclusion] Polysaccharides in Hemerocallis fulva have strong antitumor activity. The mechanism may relate to enhanced of immunifaction.
