普通小麦-华山新麦草人工合成双二倍体 AFLP标记多态性分析

摘 要:【目的】为从分子水平上了解普通小麦-华山新麦草双二倍体(PHW-SA)合成过程中遗传变异情况。【方法】用195对引物对PHW-SA及其亲本普通小麦(J-11)和华山新麦草进行AFLP标记分析。【结果】J-11、华山新麦草和PHW-SA分别扩增出标记5136、3953和5637条,分别占总标记(17726条)的29.0%、22.3%和31.8%。81.8%的J-11标记和49.6%的华山新麦草标记存在于PHW-SA中。其中J-11与PHW-SA特有相同标记2484条,分别占各自标记的48.4%和44.1%;华山新麦草与PHW-SA特有相同标记246条,分别占各自标记的6.2%和4.4%;PHW-SA特有标记232条,缺失双亲共有标记66条,分别占PHW-SA总标记的4.1%和1.2%;3种材料共有相同标记1715条,分别占J-11、华山新麦草和PHW-SA总标记的33.4%、43.4%和30.4%。【结论】PHW-SA在合成过程中基因组发生了较大的遗传变异。此研究结果为PHW-SA在小麦遗传育种中的应用提供了基础理论依据。

Abstract:

Abstract:ObjectiveThis study was used to analyze the genetic variation of PHW-SA.Method195 pairs of AFLP primers were used to amplify PHW-SA and its parents J-11(common wheat) and Psathyrostachys huashanica.Result5136, 3953 and 5637 markers were amplified in J-11, Psathyrostachys huashanica and PHW-SA, respectively, accounted for 29.0%, 22.3% and 31.8% of the total (17726 banding), respectively; 81.8% markers of J-11 and 49.6% markers of Psathyrostachys huashanica were amplified in PHW-SA; there were 2484 co-markers between PHW-SA and J-11, which accounted for 48.4% and 44.1% of themselves, respectively; similarly, 246 co-markers existed between (6.2%) PHW-SA and (4.4%) Psathyrostachys huashanica; 232 new markers which did not exist in its parents were amplified in PHW-SA and 66 markers of its parents disappeared in PHW-SA; there were 1715 co-markers in all 3 materials, accounted for 33.4%, 43.4% and 30.4% of its total markers of J-11, Psathyrostachys huashanica and PHW-SA, respectively.ConclusionAll evidence suggesting that great genetic variation was occurred in genome in the synthesize process of PHW-SA, and providing the theoretical information for wheat breeding in future.
