黑麦属NBS类抗病基因同源序列的分离与鉴定

摘要: 根据已知植物抗病基因(resistance gene, R gene)编码的蛋白质NBS-LRR保守结构设计了10对特异简并引物,对5份黑麦材料的基因组DNA进行抗病基因同源序列克隆,共获得45条抗病基因同源序列(resistance gene analogs, RGAs)。同源性比较发现,这45条RGAs均属于NBS-LRR类抗病基因类似序列,RGAs序列之间的核苷酸相似性是41.9%-99.6%,其中16条具有连续开放阅读框(ORFs)。同时,利用MEGA 3.1将这16条序列与4个已知R基因进行聚类分析,发现有13条序列与RPM1亲缘关系近,2条与Pm3a亲缘关系近。本研究在黑麦中获得的RGAs既可进一步用于筛选黑麦的抗病候选基因,同时也可以作为分子标记,并为辅助选择育种提供研究基础,且对了解黑麦中NBS-LRR类R基因的起源和进化有一定的参考价值。

Abstract: The resistance gene analogs (RGAs) from 5 Secale L. accessions were amplified using 10 pairs of specific degenerated primers based on the conserved regions of nucleotide binding site-leucine rich repeat (NBS-LRR) domain of plant resistance genes (R genes). A total of 45 RGAs were isolated, and they all belonged to NBS-LRR RGAs. The nucleotide sequences of the 45 RGAs showed the similarities of 41.9% to 99.6%. 16 out of 45 RGAs had the uninterrupted open reading frames (ORFs). 13 out of 16 RGAs with RPM1 had near relation, 2 RGAs were close to Pm3a. These results could provide some theoretic and experimental data for the isolation of related resistance genes from Secale, and the development of molecular markers for MAS. And also, it would give some information for the origin and evolution of NBS-LRR type R gene.
