Protective effects of dietary sodium selenite on the activationof death receptor pathway in thymus of the chicks exposed to AFB1

摘要: 为探究硒对AFB1致胸腺过度凋亡的拮抗机制,180只1日龄科宝肉鸡随机分为4组,分别饲以对照日粮、AFB1日粮(0.6 mg/kg AFB1)、+Se日粮(0.4mg/kg Se)和AFB1+Se日粮(0.6 mg/kg AFB1+0.4mg/kg Se)21天。结果显示,日粮硒可缓解AFB1所致的胸腺形态学损伤、T淋巴细胞亚群比例下降及凋亡率升高;与AFB1组相比,AFB1+Se组胸腺TNF-α、caspase-8、caspase-3、caspase-9和JNK1的表达量降低,Bcl-2和tBid的表达量升高。因此,日粮硒拮抗AFB1致雏鸡胸腺细胞凋亡率升高的机理与死亡受体通路中Fas-FasL- FADD-caspase8-caspase3途径和Fas-ASK1-JNK-Bcl-2途径受到干预有关。

Abstract: This experiment was conducted to investigate the protective mechanism of selenium on AFB1-induced thymocytes apoptosis. One hundred and ninety COBB chickens were randomly divided into four groups, and fed on control diet, aflatoxin B1 diet, +Se diet and AFB1+Se diet respectively for 21 days. Thymus was sampled for detecting the relative weight, T-cell subsets, percentage of apoptotic cells and relative mRNA expression of genes in the death receptor pathway at 7, 14 and 21 days of age. Our results showed that compared with the AFB1 group, Se could increase the relative thymus weight, alleviate histological lesions, reduce the incidence of apoptotic thymocytes, decrease the relative mRNA expression of JNK, TNF-α, caspase-8, caspase-3 and caspase-9, and increase the relative mRNA expression of Bid and Bcl-2. It was speculated that Se could alleviate the AFB1-induced lesions in thymus partly via the break in the following two pathways, Fas-FasL-FADD-caspase8-caspase3 pathway and Fas-ASK1-JNK-Bcl-2 pathway.
