痒螨病患兔皮损中细胞因子IFN-γ、IL-4和转录因子T-bet、GATA-3的表达

Expression of cytokine factors IFN-γ, IL-4 and transcription factors T-bet, GATA-3 in psoroptic mange with different lesion scores in rabbits

摘要: [目的] 为了探讨不同病变评分痒螨病患兔皮损中细胞因子IFN-γ、IL-4和转录因子T-bet、GATA-3的表达。[方法] 采用免疫组化、酶联免疫吸附试验和荧光定量PCR法检测痒螨病患兔(病变评分为1分、3分和5分)耳部皮损和健康兔(0分)耳部皮肤中IFN-γ/IL-4和T-bet/GATA-3的表达。[结果] 与0分组相比,患病组(1分、3分和5分组)皮损处IFN-γ蛋白和IFN-γ mRNA均增高,但仅3分组呈显著增高(IFN-γ蛋白:0分组= 0.3442 ± 0.01433 pg/mg,3分组= 0.4636 ± 0.014948 pg/mg, P < 0.05;IFN-γ mRNA:3分组/0分组=2.365,P < 0.05)。患病组皮损处T-bet和GATA-3蛋白的阳性产物均定位于细胞核,但两者较健康组均无明显差异(P > 0.05);1分组和3分组皮损中的T-bet mRNA较健康组呈显著增高(P < 0.05),而3分组和5分组皮损中GATA-3 mRNA呈显著降低(P < 0.05)。[结论] 随着痒螨病病变评分的增加,宿主的Th2免疫反应受到抑制(GATA-3的表达降低),而Th1免疫反应则有所增强(IFN-γ和T-bet的表达增高)。

Abstract: [Objective] The purpose of this study was to investigate the expression of cytokine factors IFN-γ/IL-4 and transcription factors T-bet/GATA-3 in psoroptic mange with different lesion scores. [Method] The expression of IFN-γ/IL-4 and T-bet/GATA-3 in ear skin samples of rabbits among psoroptic mange groups (lesion scores 1 group, score 3 group and score 5 group) and healthy control group (score 0 group) were investigated by immunohistochemistry, ELISA and real-time RT-PCR. [Results] Compared to score 0 group, the level of IFN-γ protein and IFN-γ mRNA increased in psoroptic mange groups (score 1 group, score 3 group and score 5 group), and there were significant differences between score 0 group (IFN-γ protein: 0.3442 ± 0.01433pg/mg) and score 3 group (IFN-γ protein: 0.4636 ± 0.014948pg/mg, IFN-γ mRNA: score 3 group/ score 0 group = 2.365; P < 0.05). Both T-bet and GATA-3 were founded in the nucleus, but there were no significant difference in the levels of T-bet protein and GATA-3 protein between psoroptic mange groups and healthy group (P > 0.05). However, compared with score 0 group, the level of T-bet mRNA was significantly increased in the score 1 and score 3 lesion groups (P < 0.05), and the level of GATA-3 mRNA was significantly decreased in the score 3 and score 5 lesion groups (P < 0.05). [Conclusion] Our results indicated that psoroptic mange rabbits elicited an inhibition of Th2 immune response and enhancement to Th1 immune response with the increases of the skin lesion scores, showing the levels of a decrease of GATA-3 and an increase of IFN-γ and T-bet.
