The effect of compound traditional Chinese medicine on return milk and lactation correlation factors in dairy cows

摘要: 【目的】探究复方中药对奶牛回乳效果及回乳期奶牛血清泌乳相关因子胰岛素样生长因子1(IGF-1)、信号转导和转录激活因子5(STAT5)和转化生长因子β1(TGF-β1)含量的影响。【方法】选取规模化奶牛场妊娠后期,即将干乳,产奶量为15.42±0.71kg的健康奶牛80头。随机分为A、B、C、D组,每组20头,采用逐渐干奶法回乳,回乳开始当天记为第0天,A组为对照组,不饲喂复方中药,B、C、D组均从回乳第1天开始,每日上午8:00分别饲喂复方中药400g、500g、600g,回乳完成后停止饲喂复方中药。分别采集回乳期奶牛第0、1、3、5、7、9和11天尾静脉血,并记录奶牛单日产奶量,ELISA法检测回乳期奶牛血清IGF-1、STAT5和TGF-β1含量。【结果】回乳期各组奶牛产奶量均随时间推移降低,A、B、C、D组回乳时间分别为11、7、5、5天。在奶牛回乳期,各组奶牛血清IGF-1和STAT5含量均随时间推移呈下降趋势,并在回乳期后回升,而各组奶牛血清TGF-β1含量均呈上升趋势,并在回乳期后下降;回乳期奶牛产奶量与血清IGF-1和STAT5含量均分别呈极显著正相关(P<0.01),与TGF-β1含量呈极显著负相关(P<0.01);回乳期奶牛血清IGF-1与STAT5含量呈极显著正相关(P<0.01),而TGF-β1与IGF-1、STAT5含量分别呈极显著负相关(P<0.01)。【结论】复方中药能明显促进回乳期奶牛回乳,饲喂500g/日即可达到最佳回乳效果。在奶牛回乳期,IGF-1和STAT5是奶牛回乳的负调控因子,而TGF-β1是奶牛回乳的正调控因子。

Abstract: 【Objective】In order to explore the impact of compound traditional Chinese medicine on the effect of return milk and serum insulin-like growth factor 1 (IGF-1), Signal Transducers and Activators of Transcription 5(STAT5) and transforming growth factor β1 (TGF-β1) levels in dairy cow during the period of milk with drawal.【Methods】80 healthy Chinese Holstein cows that in late pregnancy and gave 15.42±0.71 kg milk per day were used. The cows were randomly divided into four groups: group A, B, C and D, with 20 of them in each group. Stop dairy cows’ milk by quickly dry milk method and the day when cows begun to return milk was recorded as 0 d. Group A was a control group, did not feed compound traditional Chinese medicine and the group B, C and D were fed 400g, 500g, 600g compound traditional Chinses medicine respectively at 8.00 am from 1 d to the day of drying milk completely. We collected cows’ venous blood from the tail on the days 0, 1, 3, 5, 7, 9 and 11 d and noted the daily milk production. ELISA was used to evaluate the expression levels of serum IGF-I, STAT5 and TGF-β1.【Result】During the period of milk with drawal, the milk production of all 4 groups was on the decline Group A, B, C and D returned milk on the 11 d, 7 d, 5 d and 5 d respectively. The expression levels of serum IGF-1 and STAT5 had downward trend and raised in all 4 groups, but the expression levels of TGF-β1 had rising trend and declined after the period of milk with drawal. There were highly significant positive correction of milk production, IGF-1 and STAT5(P<0.01). The milk production showed highly significant positive correlation between the expression levels of IGF-1 and STAT5 and highly significant negative correlation between the expression level of TGF-β1(P<0.01). There were highly significant positive correction between IGF-1 and STAT5(P<0.01), while there were highly negative correction between TGF-β1 and IGF-1, STAT5. 【Conclusion】Compound traditional Chinses medicine can help cows drying milk significantly, feeding 500g/d can make drying milk to achieve the best results .During the period of milk with drawal, IGF-1 and STAT5 are negative regulation factors of returning milk, while TGF-β1 is a positive regulation factor.
