云南峨毛茶亲缘关系的ISSR分析及指纹图谱构建

ISSR Analysis on the Relationship and Construction of Fingerprint in Emao Teas of Yunnan

摘要: [目的] 了解和保护峨毛茶大茶树资源。[方法] 采用ISSR技术对云南峨毛地区的5份茶样资源的遗传多样性和亲缘关系进行研究。[结果] 结果表明:5份峨毛茶样共扩增出94条清晰的DNA条带,其中多态性带73条,占77%,遗传相似系数介于0.45~0.64之间,表明来自同一地区的峨毛茶之间已经产生遗传分化。[结论] 利用823引物构建了5份峨毛茶的指纹图谱,为今后峨毛茶的鉴定提供了参考。

Abstract: [Objective] The aim of the study was to know and protect the large tree resources of Emao teas. [Method] The genetic diversity and genetic relationship of five Emao tea samples from Yunnan were analyzed by ISSR technology. [Results] The results showed that 94 clear DNA bands in Emao tea were obtained through amplification. 73 bands of them were polymorphic and the average polymorphism rate was 77%. The genetic similarity coefficient was between 0.45 and 0.64, indicating that there was genetic diversity in Emao teas. [Conclusion] The fingerprint of five teas was constructed using the ISSR primer of 823, which can provide a molecular biology method for the identification of Emao teas.
