根癌农杆菌介导的芥菜型油菜高效遗传转化

Efficient Agrobacterium-mediated Transformation Protocol of Brassica juncea

摘要: [目的] 建立芥菜型油菜(Brassica juncea)高效遗传转化体系。[方法] 以芥菜型油菜带柄子叶为材料,采用农杆菌介导的转化方法,将外源基因导入芥菜型油菜愈伤组织,并获得再生植株。[结果] 外植体在侵染前,预先在含2mg/L 6-BA+0.5mg/L NAA+6mg/L AgNO3的MS培养基上进行预培养2~3d。侵染6~8min后转入共培养培养基中2~3d,之后转到含10mg/L卡那霉素的MS筛选培养基上进行筛选培养2~3周。对成活植株进行GUS染色和PCR验证,证明外源基因已经成功转入芥菜型油菜的基因组中。[结论] 通过优化愈伤组织诱导条件和农杆菌侵染时间,可显著提高芥菜型油菜的遗传转化效率。

Abstract: [Objective] To establish an efficient genetic transformation system of Brassica juncea.[Method] The exogenous gene was transfered into Brassica juncea's calli,cotyledonary petiole as the material for calli,by Agrobacterium-mediated transformation and regeneration plants were obtained.We optimized the Agrobacterium-mediated transformation procedure of Brassica juncea by studying the callus induction of cotyledonary petiole and plant growth regulators.[Results] The cotyledonary petiole explants were precultured on MS medium containing 2mg/L 6-BA,0.5mg/L NAA,and 6mg/L AgNO3 for 2-3days,and then were infected by A.tumefaciens,followed by co-culture for 2days,and then were transferred to selection medium containing 10mg/L kanamycin for 2-3weeks.GUS staining in transgenic plantlets confirmed the transformation of foreign DNA into plant genome.[Conclusion] The genetic transformation efficiency could be markedly improved by optimizing callus induction condition and Agrobacteriuminfection time.
