用间接免疫荧光技术检测水稻细菌性条斑病菌的研究

INDIRECT IMMUNOFLUORESCENCE TEETS FOR DETECTION AND IDENTIFCATION OF XANTHOMONAS PV. ORYZICOLA IN NATURALLY INFECTED RICE SEED

摘要: 本文报道了用于检测Xanthomonas campestris pv. oryzicola的间接免疫荧光技术。用兔抗X. c. pv. oryzlcola IgG处理标本,用羊抗兔免疫球蛋白荧光抗体染色,在荧光显微镜下观察。来自四川7个有代表性的X. c. pv. oryzicola菌株均为阳性反应,检测灵敏度为103~104个/ml。供试的X. c. pv. oryzae和X. 1eersiae为阴性反应。检测四川40份稻种,选14份用间接法酶联吸附试验进行验证,结果趋向一致,证明间接法免疫荧光技术可以快速准确地检测稻种中的X. c. pv. oryzicola。

Abstract: Indirect immunofluoresence was evaluated for detecting and identifying Xanthomonas campestris pv. oryzicola in pure culture and in rice seed. Cells were treated with immunoglobulin G, stained with fluorescein isothiocyanate and conjugated and antirabbit globulin. HF was specific for 7 X. c. pv. oryzicola isolates from 69 X. c. pv. oryzicola isolates shas were collected from diverse geographical location in Sichuan, and positive crossreacting fluorescence was not observed when IgG was tested against 3 X. c. pv. oryzae and 1 X. leersiae isolate. Cells of X. c. pv. oryzicola were detected on prepared slides in pure bacteria population containing as many as 103-104 cells per mitiliter of X. c pv. oryzicola. HF prorides a reliable method for rapid screening of rice seed lots for X. c. pv. oryzicola.
