淡紫拟青霉液体发酵条件的优化

Optimal Cultivation for Liquid Fermentation of Paecilomyces lilacinus

摘要: [目的] 对食线虫真菌淡紫拟青霉液体培养条件进行优化,从而获得优化的淡紫拟青霉液体发酵条件。[方法] 研究淡紫拟青霉在不同碳源、氮源、无机盐、初始pH值下对产孢量的影响,确定了适宜的培养条件及发酵培养时间。[结果] 最佳基础培养基配方为A1(g/L):葡萄糖40,NaNO3 20,KH2PO4 0.5,MgSO4·7H2O 0.5;最佳复合培养基配方为b(g/L):葡萄糖40,NaNO3 10,(NH4)2SO4 5,黄豆粉5,KH2PO4 0.5,MgSO4·7H2O 0.5;其他培养条件的选择结果:淡紫拟青霉最适产孢的初始pH值为6.0~7.0,发酵温度26℃,发酵培养时间为72h到80h。[结论] 通过这些研究获得了淡紫拟青霉液体优化的发酵条件,为该菌大规模产生奠定基础。

Abstract: [Objective] Our aim was to optimize the culture conditions of Paecilomyces lilacinus liquid of enmatophagous fungus, and to obtain the optimal conditions of liquid fermentation of P. lilacinus. [Method] P. lilacinus liquid was cultured under different carbon sources, nitrogen sources, inorganic salts, pH values and times. The amount of spores in each treatment was measured. Optimal condition and time were selected. [Results] ① The optimal basic medium A1(g/L):glucose 40, NaNO3 20, KH2PO4 0.5, MgSO4·7H2O 0.5;② The optimal compound medium b(g/L):glucose 40, NaNO3 10, (NH4)2SO4 5, soybean powder 5, KH2PO4 0.5, MgSO4·7H2O 0.5;③ The best initial pH value was 6.0~7.0 and temperature was 26℃;④ The optimum time for fermentation was 72~80 h. [Conclusion] This study optimizes the culture conditions of P. lilacinus and provides foundation for large scale production of P. lilacinus.
