兰尼定受体基因(RYR1)在藏猪背最长肌组织中的差异表达

Developmental Expression Differences of RYR1 Gene in Longissimus dorsal Muscle of the Tibetan Pigs

摘要: 兰尼定受体(rynodine receptor 1,RYR1)基因又称氟烷基因,已证明为猪应激综合征发生的主效基因。该基因的表达常常和猪肉质性状劣变相关联,因而具有较重要的研究前景。为了研究该基因在个体不同发育时期表达的规律性变化,为进一步深入研究该基因和肉质性状间的关系,本研究采用实时荧光定量PCR方法检测了藏猪背最长肌组织中RYR1基因在2、4、6和8月龄间表达为mRNA的量的差异。结果表明:不同月龄藏猪背最长肌中RYR1基因mRNA的表达量波动起伏,2月龄时表达量最高,4月龄表达量较2月龄极显著下降(P<0.01),6月龄又极显著上升(P<0.01),至8月龄极显著降低到最低水平(P<0.01)。本文初步揭示了藏猪RYR1基因表达在不同发育时期的变化,为进一步研究RYR1基因提供了基础数据。

Abstract: Rynodine receptor(rynodine receptor 1, RYR1) gene, also known as the halothane gene, which has been proved the major gene induced the porcine stress syndrome.The expression of the gene were associated with the pork quality traits deterioration, and thus has a more important research prospects.To investigate the regular changes of the gene expression for individuals in different developmental stages and further study the relationship between the gene and the meat quality traits, the real-time quantitative PCR was applied to test the developmental expression changes of RYR1 gene at the age of 2, 4, 6 and 8 months in Longissimus dorsal muscle of the Tibetan pigs.The results indicated that the expression level of RYR1 gene had been fluctuating at different ages, and which was highest expression at the age of 2 months, but decreased significantly(P<0.01) at 4 months compared with 2 month old, however, highly significant increased at 6 months old(P<0.01), and reduced significantly(P<0.01) to the lowest level at 8 months.These results initial revealed the expression patterns of RYR1 gene and provided some basic data for further study RYR1 gene.
