基于ISSR分子标记的红豆草资源遗传多样性分析

Genetic Diversity Analysis of Sainfoin Resources Based on ISSR Molecular Markers

摘要: 目的: 分析评价红豆草(Onobrychis viciaefolia)种质资源间亲缘关系的远近,为红豆草的良种选育提供了科学依据。方法: 利用ISSR分子标记技术对国内外22份红豆草种质资源进行遗传多样性分析。结果: 用13条引物扩增共获得101条谱带,其中多态性条带98条,多态性比率为97.12%,平均每条引物扩增出7.8个条带,平均多态性条带为7.5条,平均等位基因数(Na)、平均有效等位基因数(Ne)、Nei’s基因多样性指数(H)和香农多样性指数(I)均值分别为1.97、1.49、0.30和0.46。结论: 遗传相似性系数聚类分析表明,22份红豆草种质资源在遗传系数0.69处可聚为4大类群,其中2668号和1号单独聚为一类,与其他材料遗传差异较大。

Abstract: 【Objective】 This study aim to analyze and evaluate the genetic relationships among the germplasm resources of Onobrychis viciaefolia,and finally to provide a scientific basis for the breeding of better varieties of Onobrychis viciaefolia. 【Method】 Genetic diversity of 22 germplasm resources of red bean grass at home and abroad was analyzed using ISSR molecular markers. 【Result】 A total of 101 bands were detected by amplification using 13 markers,among which 98 were polymorphic,with a polymorphism ratio of 97.12%. An average of 7.8 bands were amplified by each primer,with an average of 7.5 polymorphic bands. The average number of alleles (Na),average effective number of alleles (Ne),Nei’s gene diversity index (H) and Shannon diversity index (I) were 1.97,1.49,0.30 and 0.46,respectively. 【Conclusion】 The clustering analysis of genetic similarity coefficient showed that the 22 germplasm resources could be clustered into four groups at the genetic coefficient of 0.69,among which No. 2668 and No. 1 were clustered into one group,which had great genetic difference from other materials.
