紫色象草辐射诱变M2代的遗传和表型变异研究

Genetic and Phenotypic Variation of Radiation Induced M2 Generation in Purple Elephant Grass

摘要: 【目的】 探究辐射诱变紫色象草M2代矮化突变体的遗传效应。【方法】 前期利用60Co-γ射线对象草种茎进行照射处理后出现了矮化的M0代突变体植株,随后通过对M0代突变体进行两年的矮化后代选择,得到了M2 代矮化植株,并对其表型性状变异和分子水平上的遗传变异进行研究。【结果】 M2代象草群体中有两株明显矮化的植株,其分蘖数、茎节数和株高与未经辐射处理的M2代材料差异显著,发生了明显的变异。SSR分子标记结果表明,在象草辐射M2代材料中,筛选出的10对引物共扩增出了209条带,其中多态性条带有201条,平均多态信息含量为0.25,突变体材料与对照之间的遗传相似系数在0.61~0.78范围内,平均值为0.71。UPGMA聚类结果显示矮化突变体 M2-8和M2-17聚为一类,与对照遗传距离较远,变异程度较大,可为将来的象草育种项目中提供原始材料。【结论】 矮化突变体的突变原因很大可能性是由遗传因素所引起的变异。

Abstract: 【Objective】 To investigate the genetic effect of radiation induced M2 dwarfing mutant of Purple elephant grass. 【Method】 In the early stage, our team irradiated the stem of grass with 60Co-γ rays, and dwarf M0 generation mutant plants appeared. Then, M2 generation dwarf plants were obtained through the selection of dwarf offspring of M0 generation mutants for 2 years, and the phenotypic trait variation and genetic variation at the molecular level were studied. 【Result】 The results illustrated that there were two dwarfed plants in the M2 generation. The number of tillers, nodes and plant height were significantly different from those in the M2 generation without radiation treatment, and there were obvious variations. The results of SSR molecular markers showed that a total of 209 bands were amplified from the 10 pairs of primers screened out in the M2 generation of the weaselgrass radiation material, among which 201 were polymorphic bands. The average polymorphism information content was 0.25. The genetic similarity coefficient between the mutant material and the control material was in the range of 0.61-0.78, with an average of 0.71. UPGMA clustering results manifested that the dwarf mutants M2-8 and M2-17 for a class, and contrast heredity distance is farther, variation is larger, but for the future of elephant grass breeding project provides raw materials. 【Conclusion】 The mutation of dwarfing mutants is probably caused by genetic factors.
