三七丹参片对高血脂小鼠血脂水平的影响

Effect of Sanqi Danshen Tablet on Blood Lipid Level of Hyperlipidemic Mice

摘要: 【目的】 采用饲喂高脂饲料方法建立稳定的昆明小鼠高脂血症模型,并使用该模型小鼠对三七丹参片降血脂作用进行研究。【方法】 将80只小鼠分为模型组(20只,雌雄各半)和降脂组(60只,雌雄各半)。模型组随机分为正常组和高脂组(n=10),雌雄各半;正常组给予普通饲料喂养,高脂组给予高脂饲料喂养,连续饲喂30 d。计算增重率;检测总胆固醇、三酰甘油、高密度脂蛋白和低密度脂蛋白等指标;比较肝脏组织病理变化。降脂组随机分为空白对照组,高脂模型组,阳性对照组(辛伐他汀组),三七丹参片低、中、高剂量组共6组(n=10),雌雄各半。对各组小鼠灌服三七丹参、辛伐他汀和生理盐水。连续灌胃 30 d,每天称重。检测血脂和血糖的变化情况。【结果】 与对照组相比,高脂组小鼠血脂4项指标结果显示TC、TG、LDL-C显著升高(P<0.01或P<0.05),HDL-C差异无统计学意义(P>0.05);高脂组肝细胞均出现脂肪样变性。使用降脂药后,高剂量组的血脂和血糖指标均降低。【结论】 试验建立了稳定的高血脂小鼠模型,三七丹参片对高脂食物造成的高血脂小鼠有很好的降血脂和降血糖作用。

Abstract: 【Objective】 To establish a stable animal model of hyperlipidemia in Kunming mice with high-fat diet. 【Method】 80 Kunming mice were divided into model group (20 mice, half male and half female) and antihypertensive group (60 mice, half male and half female). The model group was randomly divided into normal and high fat groups (n=10); The normal group was fed with normal diet, and the high-fat group was fed with high-fat diet. They were all fed for 30 days. The antihypertensive group was randomly divided into blank control, high-fat model, positive control (simvastatin), low, medium and high dose groups of Sanqi Danshen Tablets groups (n=10). Mice in each group were perfused with Panax notoginseng, Salvia miltiorrhiza, simvastatin and normal saline for 30 consecutive days. The weight gain rate was calculated. Blood samples were collected and the total cholesterol, triglyceride, high density lipoprotein and low density lipoprotein were detected. The pathological changes of liver tissue and the blood lipid and blood glucose in each group were observed. 【Result】 Results showed that compared with the control group, TC, TG and LDL-C increased after modeling in fat emulsion group (P<0.01 or P<0.05), but HDL-C had no significant difference (P>0.05). After drugs used, blood fat and blood glucose indexes of the high-dose group was decreased. 【Conclusion】 The stable hyperlipidemia mice model was established. Sanqi Danshen Tablet had good hypoglycemic and hypolipidemic effects on hyperlipidemia mice caused by high fat food.
