HINT1基因多态性及其在肌肉组织中表达差异的分析

Analysis of HINT1 Gene Polymorphism and Its Expression Difference in Muscle Tissue of Pig

摘要: 【目的】 探究HINT1基因在猪肌肉生长分子调控机制中的作用。【方法】 试验选取30、90和180日龄的藏猪和大约克猪(各8头)作为试验对象,利用实时荧光定量PCR技术检测HINT1基因在背最长肌、腿肌组织中mRNA的差异表达,采用Sanger测序法对藏猪(48头)和大约克猪(45头)HINT1基因的起始密码子上游3 000 bp区域左右进行单核苷酸多态性分析。【结果】 在背最长肌和腿肌组织中,藏猪(TP)和大约克猪(YY)HINT1基因mRNA的表达量均随着日龄的增长呈逐渐降低的趋势;在背最长肌组织中,30日龄藏猪表达量极显著高于大约克猪(P<0.01),90日龄藏猪和大约克猪表达差异不显著(P=0.284),180日龄藏猪显著高于大约克猪(P<0.05);在腿肌组织中,30日龄和90日龄的藏猪HINT1基因表达量均极显著高于大约克猪(P<0.01),180日龄藏猪的表达量显著高于大约克猪(P<0.05);在起始密码子上游3 000 bp区域左右筛选到5个SNPs位点:A-2230G、C-2209T、A-2174G、T-2083C和A-2082G ,且两猪种间等位基因频率存在极显著差异(P<0.01),品种内基因频率均符合哈迪温伯格定律。【结论】 推测HINT1基因的表达与骨骼肌组织的生长发育呈负相关。研究结果为进一步改善猪肉质品质及调控猪肌肉组织生长发育的分子机制提供了理论依据。

Abstract: 【Objective】 To explore the role of HINT1 gene in the molecular regulation mechanism of pig muscle growth. 【Method】 Tibetan pigs and Yorkshire pigs (8 pigs respective) aged 30, 90 and 180 days were selected as experimental subjects, Real-time quantitative PCR was used to detect the mRNA differential expression of HINT1 gene in longissimus dorsi muscle and leg muscle tissue. Single nucleotide polymorphism analysis was performed in the upstream 3 000 bp region of the start codon of HINT1 gene of Tibetan pigs (n=48) and Yorkshire pigs (n=45) with the Sanger sequencing method. 【Result】 The results showed that the mRNA expression of HINT1 gene in longissimus dorsi muscle and leg muscle of Tibetan pig (TP) and Large Yorkshire (YY) decreased with the increase of age; In longissimus dorsi muscle tissue, the expression level in HINT1 gene of Tibetan pigs at 30 days of age was significantly higher than that in Yorkshire pigs (P<0.01), and there was no significant difference between Tibetan pigs and Yorkshire pigs at 90 days of age (P=0.284), but the expression level of Tibetan pigs at 180 days of age was significantly higher than that in Yorkshire pigs (P<0.05). In the leg muscle tissue, the expression level of HINT1 gene in 30-day-old and 90-day-old Tibetan pigs was extremely significantly higher than that in Yorkshire pigs (P<0.01), and the expression level of 180-day-old Tibetan pigs was significantly higher than that in Yorkshire pigs (P<0.05). Five SNPs were screened in the upstream 3 000 bp region of the starting codon: A-2230G, C-2209T, A-2174G, T-2083C and A-2082G, and there were significant differences in allele frequencies between the two breeds (P<0.01). The gene frequencies within the breeds were in accordance with Hardy-Weinberg equilibrium. 【Conclusion】 It is speculated that the expression of HINT1 gene is negatively correlated with the growth and development of skeletal muscle tissue. The results of this study provide a theoretical basis for further improving pork quality and regulating the molecular mechanism of pig muscle tissue growth and development.
