无患子叶斑病病原菌鉴定及其生物学特性

Identification and Biological Characteristics of Pathogen Causing Leaf Spot Disease of Sapindus mukorossi

摘要: 【目的】 明确福建顺昌无患子叶斑病病原及其生物学特性。【方法】 以福建省顺昌县受害无患子人工林基地采集的病样组织为研究对象,通过组织分离法、致病性测定、形态学鉴定及分子生物学鉴定,确定无患子叶斑病病原菌种类,在此基础上探究病原菌生物学特性。【结果】 通过组织分离法从发病部位分离出9种菌株,经柯赫氏法则验证,确定SW-9为无患子叶斑病病原菌。结合传统形态学和分子生物学,将该菌鉴定为Xylaria arbuscula(矮乔木炭角菌)。SW-9菌株最适生长温度为28 ℃;最适生长pH为6~7;最适碳源为麦芽糖,最适氮源为蛋白胨;其致死温度为40 ℃;最适光照条件为全黑暗培养。【结论】 关于Xylaria arbuscula引起的无患子叶斑病为首次报道,研究结果将为开展无患子叶斑病的防治工作奠定基础。

Abstract: 【Objective】 To clarify the pathogen causing leaf spots of Sapindus mukorossi in Shunchang County and its biological characteristics. 【Method】 Taking the disease sample tissue collected from the Sapindus mukorossi artificial forest base in Shunchang County, Fujian Province as the research object, to determine the types of pathogen by tissue separation, pathogenic determination, morphological identification and molecular biological identification, and explore the biological characteristics of the pathogen. 【Result】 Nine species of fungi were isolated from the site by tissue separation, SW-9 was identified as pathogen of leaf spot disease of Sapindus mukorossi. Combining traditional morphology and molecular biology, the fungus was identified with Xylaria arbuscula. The most suitable growth temperature is 28 ℃; most suitable growth pH is 6-7; most suitable carbon source is maltose, and most suitable nitrogen source is protein; the death temperature is 40 ℃; most suitable light conditions are all dark culture. 【Conclusion】 The leaf spot disease of Sapindus mukorossi caused by Xylaria arbuscula is the first reported, and the results will lay the foundation for the prevention and treatment of the leaf spot disease of Sapindus mukorossi.
