水柏枝乙醇提取物不同极性部位体外抗氧化活性研究

Study on the Antioxidant Activity in vitro of Different Polar Parts of Ethanol Extract of Myricaria gerynanica(L.) Desv.

摘要: 【目的】探究水柏枝乙醇提取物不同极性部位的体外抗氧化活性。【方法】以水柏枝的70%乙醇提取物为样品,对醇提物进行萃取,得到水柏枝醇提物的不同极性部位并对其进行大类成分鉴别;用Folin-Ciocalteu方法和氯化铝比色法分别测定总多酚和总黄酮的含量;采用DPPH、ABTS和总还原能力3个指标来全面评估其体外抗氧化活性。【结果】初步植物化学筛选结果表明,水柏枝醇提物中不同程度地含有多酚、黄酮、香豆素、甾体和糖苷等类化合物,其中黄酮类在水柏枝的各个极性部位均有分布,酚类主要分布在乙酸乙酯和正丁醇部位,皂素主要分布于二氯部位,香豆素类主要集中在乙酸乙酯部位;萜类以及甾体主要分布在石油醚二氯部位。多酚和黄酮类含量测定结果表明,水柏枝不同极性部位当中,乙酸乙酯部位的多酚含量最高(94.43±1.5) mg/g,乙酸乙酯部位中总黄酮含量最高(627.21±1.2) mg/g。DPPH、ABTS和总还原能力3个指标综合得出水柏枝醇提物的乙酸乙酯部位的抗氧化能力最佳,正丁醇部位抗氧化能力次之。【结论】水柏枝乙醇提取物不同极性部位均有一定的抗氧化活性,其中乙酸乙酯部位活性最佳,且优于BHT,其良好的抗氧化能力可能与其中高含量的多酚和黄酮成分相关。

Abstract: 【Objective】 To explore the antioxidant activity of different polarity part of ethanol extract from Myricaria gerynanica (L.) Desv. in vitro. 【Method】 Taking the 70% ethanol extract of Myricaria gerynanica (L.) Desv. as a sample, the different polar parts were obtained by extraction with different solvent, which were identified the main chemicals; Folin-Ciocalteu method and aluminum chloride colorimetric method were used to determine the content of polyphenols and flavonoids. Three indicators, DPPH, ABTS, and total reducing capacity, were used to evaluate its anti-oxidant activity in vitro. 【Result】 Preliminary phyto-chemical screening results showed that the ethanol extract of Myricaria gerynanica (L.) Desv. contains polyphenols, flavonoids, coumarins, steroids, glycosides and other compounds to vary degrees, and the flavonoids are distributed in all polar parts of Myricaria gerynanica (L.) Desv., phenols are mainly distributed in ethyl acetate and n-butanol, saponins are mainly distributed in dichloride part, coumarins are mainly concentrated in ethyl acetate; terpenoids and steroids are mainly distributed in petroleum ether dichloride part. The results of determination of phenol and flavonoids content showed that the ethyl acetate part has the highest contents of total polyphenols (94.43±1.5) mg/g and total flavonoids (627.21±1.2) mg/g. The ethylacetate part exhibited the best antioxidant capacity in DPPH,ABTS, and total reduction capacity tests, then followed by n-butanol part. 【Conclusion】 All the different polar parts of the ethanol extract from Myricaria gerynanica (L.) Desv. exhibited certain antioxidant activity. Among them, the ethyl acetate part has the best antioxidant activity, better than B HT. Its good antioxidant capacity may own to the high content of polyphenols and flavonoids.
