山苍子精油抑制沙门菌作用机制研究

Antibacterial Mechanism of Litsea cubeba Essential Oil on Salmonella

摘要: 【目的】探究山苍子精油对沙门菌的抑菌机制。【方法】采用二倍稀释法和平板计数法测定最小抑菌浓度(MIC)和最小杀菌浓度(MBC);通过测定菌液电导率和细胞膜电位研究山苍子精油对细胞膜的影响;利用透射电镜和扫描电镜观察山苍子精油对细胞形态结构的影响;通过SDS-PAGE研究山苍子精油对可溶性蛋白的影响;采用结晶紫染色法探究亚抑菌浓度的山苍子精油对沙门菌生物被膜形成能力的影响。【结果】山苍子精油对鼠伤寒沙门菌的MIC和MBC均为256 μg/mL。与对照组相比,2 MIC浓度的山苍子精油作用于菌体,1 h内菌悬液电导率和胞膜电位明显上升;12 h后,菌体可溶性蛋白质含量降低(41.69±0.97)%,菌体形态结构完整性被破坏。亚抑菌浓度下,经8 μg/mL山苍子精油处理后,生物被膜总量减少(19.25±3.42)%。【结论】山苍子精油可改变沙门菌的细胞通透性、形态结构,减少可溶性蛋白质含量和抑制生物被膜的形成,从而起到良好的抑菌和杀菌作用。

Abstract: 【Objective】 This study was aimed to investigate the antibacterial mechanism of Litsea cubeba essential oil (LC-EO) on Salmonella typhimuriumin. 【Method】 The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of LC-EO to S. typhimuriumin were determined via dilution method and colony counting method. Bacterium solution conductivity and membrane potential were detected to investigate the effect of LC-EO on the bacterial cell membrane. The effect of LC-EO on bacterial morphology was observed by transmission electron microscope and scanning electron microscopy. SDS-PAGE was used to detect the effect of LC-EO on the content of bacterial soluble proteins. The effect of LC-EO on biofilm formation was investigated via the violet staining method. 【Result】 Both MIC and MBC of LC-EO to S. typhimuriumin were 256 μg/mL. Compared with the control group, after being treating the strain with 2 MIC LC-EO, the bacterium solution conductivity and membrane potential were increased in 1 h. After being treating for 12 h, the soluble protein content of the bacteria was reduced by (41.69±0.97)% and the integrality of morphological structure was destroyed. After 8 μg/mL LC-EO treatment, the total amount of biofilm decreased by (19.25±3.42)% at the subinhibitory concentration. 【Conclusion】 LC-EO changed the cell permeability and morphological structure of Salmonella, reduced the content of soluble protein and inhibit biofilm formation, and consequently carried out antibacterial and bactericidal functions.
