冠菌素对番茄防御基因表达、胼胝质沉积及细菌生长的影响

The Effect of Coronatine on Tomato Defense Gene Expression,Callose Deposition and Bacterial Growth

摘要: 【目的】探究冠菌素(coronatine,COR)在丁香假单胞菌与番茄互作过程中的作用机理。【方法】以番茄品种Money maker为试验材料,用RT-qPCR法检测不同浓度COR注射番茄叶片之后防御基因表达量,利用苯胺蓝染色法观测叶片中不同菌株诱导的胼胝质沉积数目,采用稀释平板计数法检测细菌生长量。【结果】RT-qPCR结果显示高浓度COR处理番茄可诱导EIN2EIN3LoxDMYC2PAL5基因的高表达,抑制PR1a基因的表达;细菌生长测定结果显示野生型病原菌比COR缺失突变体在番茄叶片中的生长量高5倍;染色观测结果显示COR能够抑制番茄胼胝质的沉积。【结论】COR影响番茄激素信号途径,抑制胼胝质沉积,促进病原菌在番茄叶片中的生长。

Abstract: 【Objective】 To investigate the meachanism of coronatine (COR) function during Pseudomonas syringae and tomato interaction.【Method】 Using tomato variety Money maker as the test material,RT-qPCR was used to detect the expression of defense genes in tomato leaves after the injection of different concentrations of COR,and the aniline blue staining method was used to observe the number of callose deposits induced by different strains in leaves,using a series of dilution plate counting method detects bacterial growth.【Result】 RT-qPCR results showed that high concentration COR treatment could induce high expression of EIN2, EIN3,LoxD, MYC2, PAL5,and inhibited the expression of PR1a.the results of growth curve assays showed that wild-type pathogenic bacteria can obtain greater population density in tomato leaves than COR deletion mutants; staining observation results showed that COR could inhibit callose deposition.【Conclusion】 COR is involved in regulating the tomato hormone pathways,inhibiting callose deposition,and promoting the growth of pathogenic bacteria in tomato leaves.
