藏猪和大约克猪GHRIGF-I基因表达模式分析

Analysis of GHR and IGF-I Gene Expression Patterns in Tibetan and Yorkshire Pigs

摘要: 【目的】探究西藏高原环境下藏猪和大约克猪GHRIGF-I基因多态性和表达模式。【方法】采用Sanger测序技术对藏猪(62头)和大约克猪(61头)GHRIGF-I基因起始密码子上游3 kb区域进行多态性检测;利用RT-qPCR技术检测了30、90及180日龄藏猪和大约克猪(各8头)背最长肌组织GHRIGF-I mRNA表达模式。【结果】藏猪与大约克猪GHR基因A-513G位点基因型频率呈显著差异(P<0.05),T-2201C位点基因型频率呈极显著差异(P<0.01);IGF-I基因G-2413A位点基因型频率呈极显著差异(P<0.01)。在mRNA表达水平,GHRIGF-I基因在品种内具有相同的表达模式;在品种间,表达模式具有明显差异。【结论】这3个SNPs位点可能是影响猪生长发育的重要调控位点,为进一步阐明GHRIGF-I基因在猪生长发育过程中的分子机制奠定基础。

Abstract: 【Objective】 In order to explore the polymorphisms and expression patterns of GHR and IGF-I genes in Tibetan and Yorkshire pigs in the Tibetan Plateau. 【Method】 Sanger sequencing technology was used to detect polymorphisms in the 3 kb region upstream of the start codons of the GHR and IGF-I genes in Tibetan pigs (62) and Yorkshire pigs (61); RT-qPCR technology was used to detect the expression patterns of GHR and IGF-I mRNA in the dorsal longest muscle tissues of Tibetan pigs and Yorkshire pigs (8 pigs each group) at 30, 90 and 180 days. 【Result】 The results showed that there was a significant difference in the genotype frequency of the GHR A-513G locus between Tibetan pigs and Yorkshire pigs (P<0.05), while the genotype frequency of T-2201C locus showed an extremely significant difference (P<0.01). The genotype frequency of G-2413A locus of IGF-I gene showed a significant difference between Tibetan pigs and Yorkshire pigs (P<0.01). At the mRNA expression level, the GHR and IGF-I genes had the similar expression pattern in the Tibetan pigs and Yorkshire pigs. There are significant differences in expression patterns between Tibetan pigs and Yorkshire pigs. 【Conclusion】 It is speculated that these three SNPs might be important regulatory sites affecting pig growth and development, which lays a foundation for further elucidating the molecular mechanism of GHR and IGF-I genes in pig growth and development.
