Identification of the Possible Proteins Responsible for Celiac Disease in Potato by Mass Spectrum

摘要: 【目的】为马铃薯品质育种提供理论依据。【方法】利用小麦麸质蛋白提取液提取马铃薯储藏蛋白,然后利用十二烷基苯磺酸钠聚丙烯酰胺凝胶电泳分离可能的致敏蛋白,切胶回收目的蛋白,真空干燥,酶切后质谱检测分离的蛋白,并与小麦中已知的致敏性多肽进行比对。【结果】共检测到199种马铃薯蛋白,比对后未发现其含有可导致乳糜泻的多肽。【结论】马铃薯不含有类似麸质蛋白的多肽。

Abstract: 【Objective】 This research would provide basic knowledge for potato quality breeding. 【Method】In this experiment, wheat gluten extracting solution was used to extract storage proteins in potato, and then these proteins were separated by sodium dodecyl sulfate polyacrylamide gel elecrophoresis (SDS-PAGE). Desired bands were cut from the PAGE gel, before they were cut into pieces. Gel pieces were placed under vacuum centrifugation until completely dried, and then digested by trypsin. Mass measurements were performed on a LTQ mass spectrometer (MS). The acquired MS spectra were searched against the known wheat allergenic CD (celiac disease) epitopes. 【Results】 In total, 199 proteins were identified by MS, and none of them shown similarity to the known wheat allergenic CD peptides. 【Conclusion】 Potato does not contain any CD epitope.
