三聚氰胺致猫慢性肾衰模型的建立

Establishment of Melamine-Induced Chronic Renal Failure Model in Cats

摘要: 【目的】建立猫慢性肾衰(CRF)模型,为研究猫CRF的发病机制及评价猫 CRF 药物的疗效奠定基础。【方法】将三聚氰胺与三聚氰酸1∶1混合物按体重给予20、10、5 mg/kg剂量拌料连续饲喂试验组猫21 d(1次/d),后再按1次/2 d饲喂14 d(7次),于0、22、36 d采集血液、尿液检测生理生化指标;按照国际公认的猫慢性肾衰分级标准,判定模型建立是否成功;死亡猫剖检取肾组织制作病理切片,HE染色观察其病理学变化。【结果】①试验组猫出现多饮多尿、消瘦等肾衰临床症状,血肌酐、尿素氮升高,红细胞数目、尿比重下降,并有尿蛋白产生。②肾脏病理组织学结果显示,肾小管上皮细胞空泡变性、坏死,肾小管上皮细胞脱落在管腔中央形成肾小管上皮细胞管型,肾小球结构不完整。【结论】采用低(5 mg/kg)、中(10 mg/kg)剂量三聚氰胺与三聚氰酸1∶1混合物先每天1次连续饲喂猫21 d、再按两天1次饲喂14 d的方法能建立稳定的轻度或中度猫CRF模型。

Abstract: 【Objective】 To establish a cat model of chronic renal failure (CRF), which lay a foundation for studying the pathogenesis of CRF in cats and evaluating the therapeutic effect of cat CRF drugs. 【Method】 A 1:1 mixture of melamine and cyanuric acid was continuously fed to test group cats at 20, 10 and 5 mg/kg for 21 days (21 times), and then fed once by two days for 14 days (7 times). Collected the blood and urine on 0,22 and 36 d to measure physiological and biochemical indexes. According to internationally recog-nized cat chronic renal failure grading standards to determine whether the model is successfully esta-blished. Finally, the renal tissues of dead cats were dissected to make pathological sections, and the pathological changes were observed by HE staining. 【Result】① The cats in the test group showed clinical symptoms of renal failure such as polydipsia, polyuria, and marasmus, with increased serum creatinine and urea nitrogen, decreased red blood cell number and urine specific gravity, and urine protein production. ② The renal histopathological results showed that the tubular epithelial cells vacuolar dege-neration and necrosis, tubular epithelial cells were shed in the center of the tubular lumen to form tubular type of renal tubular epithelial cells, and the glomerular structure was incomplete. 【Conclusion】 A stable mild or moderate cat CRF model could be established by feeding a 1:1 mixture of melamine and cyanuric acid at low (5 mg/kg) and medium (10 mg/kg) doses for 21 days, and then feeding the mixture once/two days for 14 days.
