利用SSR分子标记构建名山茶树基因身份证

Construction of SSR-Based Molecular IDs for Tea Planted in Mingshan

摘要: 【目的】揭可用于精准快速鉴别四川省名山茶树良种繁育场的茶树种质资源。【方法】利用微卫星标记技术对24份名山良种繁育场具有代表性的茶树品种进行指纹鉴定和种质识别研究。【结果】供试品种间的遗传距离为0.3~0.89,在0.89可将24份材料完全区分开,其中特色茶树品种紫嫣在0.3与其他23份品种分开,单为一类,反映出该品种与其他供试材料的亲缘关系较远。使用7对引物的PCR扩增基因型排列组合,赋予一定的阿拉伯数字,构建出的分子二维码,能被众多扫码机器识破进行快速解读。【结论】构建的二维码身份证可实现对24份茶树品种精确鉴定,为名山茶树良种场种质资源鉴定与保护提供了依据。

Abstract: 【Objective】 The objective of the study was to provide an accurate and rapid identification approach for tea germplasm resources of Mingshan tea plant in Sichuan Province. 【Method】 Microsatellite marker technology was used to fingerprint 24 representative tea clonal cultivars from Mingshan of Sichuan. 【Result】 The genetic distances among the tested cultivars ranged from 0.3 to 0.89. The 24 cultivars were completely separated at a genetic similarity of 0.89. Among them,‘Ziyan’, a new cultivar with purple young shoots, was separated from the other 23 cultivars at a similarity of 0.3, reflecting that‘Ziyan’had a relative far relationship with others. According to the PCR amplification Genotype arrangement of the seven pairs of primers, a certain number of Arabic numerals were given. Meanwhile, the molecular molecular QR code can be quickly interpreted by many scanning machines. 【Conclusion】 The molecular qr code IDs based on marker genotypes is helpful for identification and protection of tea germplasm resources of Mingshan tea.
