黑小麦阿拉伯木聚糖酶提工艺优化及抗氧化活性研究

Optimization of Enzyme Extraction Process of Triticale Arabinoxylan and Antioxidant Activity

摘要: 【目的】研究黑小麦阿拉伯木聚糖(arabinoxylan,AX)的最佳酶提工艺及其抗氧化活性。【方法】以黑小麦为原料,利用响应面法优化木聚糖酶提取AX的最佳工艺条件,与纤维素酶提作对比;将两种酶提残渣用碱进行提取,比较分析酶提与残渣碱提所得AX的抗氧化性。【结果】木聚糖酶提AX(xylanase extract arabinoxylan,XAX)的最佳工艺为木聚糖酶量39.99 mg、超声时间90.20 min、超声温度56.13 ℃、超声功率150 W,AX得率为4.83%,碱提残渣XAX-1得率为11.21%;纤维素酶提AX(cellulase extract arabinoxylan,CAX)得率为2.33%,碱提残渣CAX-1得率为15.98%。XAX、CAX对·OH和DPPH的清除能力强于XAX-1和CAX-1;XAX、CAX、XAX-1、CAX-1的还原力较弱且差异不显著;铁离子螯合率为XAX-1、CAX-1>XAX、CAX,而XAX及XAX-1的O2-清除率优于CAX及CAX-1。【结论】使用碱处理酶提所得残渣可以提高AX得率;4种方式提取的AX均具有一定的抗氧化性,但其抗氧化性不完全与阿魏酸含量呈正相关。

Abstract: 【Objective】 The objective was to study the optimum enzymatic extraction process and the antioxidant activity of arabinoxylan(AX) from triticale. 【Method】 Using triticale as raw material, response surface methodology was used to select optimum process conditions for extracting AX by xylanase, compared with cellulase extraction. Two enzymed residues were extracted to obtain AX by alkaliand antioxidant activity of AXs was analyzed. 【Result】 The optimum conditions for extracting AX by the xylanase were (xylanase extract arabinoxylan, XAX) as follows, xylanase 39.99 mg, ultrasonic time 90.20 min, ultrasonic temperature 56.13 ℃ and ultrasonic power 150 W. Under the optimized conditions, the yield of AX was 4.83%, the alkali extraction residue XAX-1 yield was 11.21%, while cellulase extraction arabinoxylan (CAX) yield was 2.33% and alkali extraction residue CAX-1 yield was 15.98%. XAX and CAX had better clearance ability of OH and DPPH than XAX-1 and CAX-1. However, the reducing power of all four AXs was weak and non-significant difference. The chelation rates of iron ions were XAX-1, CAX-1>XAX and CAX and O2-· clearance rates of XAX and XAX-1 were better than those of CAX and CAX-1. 【Conclusion】 The yield of AX from triticale was increased by treating enzyme extraction residue with alkali, which realized the rational use of resources. These four AXs all possessed certain antioxidant activity, but antioxidant activity was not completely related to the content of ferulic acid.
