Cloning and Evolution Analysis of FSHR Gene in Forest Musk Deer (Moschus moschiferus)

摘要: 【目的】克隆林麝FSHR基因有助于了解其结构、功能及进化。【方法】运用PCR产物克隆测序的方法获得了FSHR基因的DNA序列序列,并运用Chmmosoma 1.62、DNAStar 7.2等生物信息学软件分析了基因的进化和相关蛋白质性质。【结果】获得一段全长为2 088 bp的林麝FSHR基因DNA序列,含有一个CDS区(1 971 bp)和5’UTR区(117 bp),共编码656个氨基酸残基(GeneBank登记号为:MG787948);蛋白为疏水性蛋白。碱基序列的同源性分析发现在偶蹄目中具有很高的同源性,林麝与家牛(Bos taurus)的相似性最高(96.1%),与原鸡(Gallus gallus)最低(71.2%)。基因进化分析显示用13个物种FSHR基因CDS序列构建的NJ树与ME树结构一致,表明FSHR基因适合用于构建不同物种间的系统进化树。【结论】林麝FSHR基因的克隆和进化分析为今后深入研究促卵泡激素受体基因功能以及开展林麝相关功能基因的表达机制研究提供了理论依据。

Abstract: 【Objective】 The full-length cDNA sequence of FSHR gene was cloned to explore the structure, function and evolution of the FSHR gene in Forest Musk Deer (Moschus moschiferus). 【Method】 The DNA sequence of FSHR gene was obtained by PCR product cloning and sequencing, and the evolution of genes and related protein properties were analyzed by bioinformatics software including Chmmosoma 1.62 and DNAStar 7.2. 【Result】 The DNA sequence of a 2 088 bp Moschus moschiferus FSHR gene containing a CDS region (1 971 bp) and a 5′ UTR region (117 bp) encoding a total of 656 amino acid residues (GeneBank accession number: MG787948) was obtained. The protein was a hydrophobic protein. The homology analysis of the base sequence revealed high homology in the genus Artiodactyla. Moschus moschiferus showed the highest similarity with Bos taurus (96.1%), and lowest with the Gallus gallus (71.2%). Gene evolution analysis showed that the NJ tree constructed with 13 species of FSHR gene CDS sequence was consistent with the ME tree structure, indicating that the FSHR gene is suitable for constructing phylogenetic trees between different species. 【Conclusion】 Cloning and evolutionary analysis of FSHR Gene in musk deer contribute to the further study the function of FSHR gene and the study of conduct the expression mechanism on musk deer related functional genes.
