菊花在形态、组织结构和基因表达方面对弱光的响应

Research of Chrysanthemum morifolium in Morphology,Tissue Structure and Gene Expression

摘要: 【目的】探索2个菊花品种清露和公子对弱光响应的差异。【方法】设定自然光照和20%自然光照2个光照条件,测量叶片形态、茎解剖结构、叶片超微结构及相关基因的表达。【结果】生长在20%光照条件的植株徒长、节间变长,比叶重下降,经双因素分析发现,品种因子对节间长和叶柄长影响达到显著水平,光照因子对节间长、叶柄长影响达到极显著水平,对比叶重影响达到显著水平。弱光条件下清露表皮和皮层细胞呈不规则变形,维管束细胞显著变形,公子受到的影响较小。在弱光处理条件下,清露和公子叶片叶绿素a/b结合蛋白基因2(CmLhcb2)和1-氨基环丙烷-1-羧酸氧化酶基因2(CmACO2)表达量均有不同程度的上调,清露叶片CmLhcb2在0~3 d内快速上调,CmACO2在第1 天显著增加,第2 天开始下降,而公子叶片中以上2个基因的表达量在0~4 d逐步增加。【结论】随着弱光处理时间的延长,清露在组织结构和超微结构方面都受到影响严重,而公子作了适应性调整来抵御弱光胁迫。

Abstract: 【Objective】 In order to explore the differential responses of two chrysanthemum morifolium cultivars Puma Sunny and Gongzi under low light. 【Method】 The cultivars were cultivated under natural light and low light (20% of natural light) treatments to assess differences in leaf morphology, stem structure, leaf ultrastructure and relevant gene expression. 【Result】 The plants cultivated under 20% natural light grew excessively. The length of internodes increased, and specific leaf weight decreased. According to the results of two-way ANOVA, the cultivar factor had considerable effects on both internode and petiole length. The effects of the light factor on internode length, petiole length, and specific leaf weight were extremely significant. Under low light treatment, the epidermis and cortical cells of Puma Sunny were deformed and irregular. Its vascular bundle cells were also significantly deformed. Comparatively, Gongzi was less affected by these factors. Under low light treatment, the chlorophyll a/b of Puma Sunny and Gongzi interacts with protein gene 2 (CmLhcb2) and 1-aminocyclopropane-1-carboxylate oxidase gene 2 (CmACO2). Their expression quantities were up regulated at different levels. For Puma Sunny, CmLhcb2 was up regulated rapidly at 0 d. The CmACO2 level increased significantly within the first day but started decreasing the next day. For Gongzi, the expression quantities of these two genes increased gradually over a period of 0-4 days. 【Conclusion】 With prolonged time under low light treatment, Puma Sunny was severely affected in terms of tissue structure and ultrastructure, whereas Gongzi was capable of resisting these changes under low light intensity.
