响应面法优化鲟鱼肝脏铁蛋白提取工艺

Optimization of Extraction Process of Ferritin from Sturgeon Liver by Response Surface Methodology

摘要: 【目的】利用鲟鱼肝脏等下脚料提取铁蛋白,并优化提取工艺。【方法】在单因素试验的基础上,根据Box-Behnken原理采用响应面分析法,选择料液比、硫酸铵饱和度、pH为自变量,铁蛋白提取率为响应值设计实验。【结果】最佳提取工艺为料液比1∶3.8,硫酸铵饱和度57%,pH 8.12。铁蛋白的得率0.003 295%,与理论值0.003 386%偏差较小。【结论】响应面法是优化鲟鱼肝脏铁蛋白提取工艺的合理方法,本研究结果具有方法学参考意义。

Abstract: 【Objective】 The aim of this study was to prepare ferritin from sturgeon liver, and to optimize the extraction process by response surface analysis. 【Method】 Based on single factor experiment, factors includingsolid-liquid ratio, ammonium sulfate saturation and pH were selected as independent variables,and ferritin extraction rate was selected as the response value. According to the principle of Box-Behnken.【Result】 The optimum extraction condition for ferritin was as follows: solid-liquid of 1:3.8, ammonium sulfate saturation of 57%, pH of 8.12. The yield of ferritin was 0.003 295%, which was close to the theoret-ical value (0.003 386%). 【Conclusion】 The response surface methodology was at least one of the effective optimization method for ferritin extraction, and the results of this study provides reference for optimizing the extraction process of liver ferritin from sturgeon.
