牛角地黄汤对金黄色葡萄球菌耐药性的影响及转录组学分析

Effect of Niujiaodihuang Decoction on Drug Resistance and the Genetic Transcription of Staphylococcus aureus

摘要: 【目的】探讨耐药金黄色葡萄菌(MRSA)在牛角地黄汤作用下对甲氧西林敏感性的影响。【方法】采用微量稀释法检测金黄色葡萄球菌在牛角地黄汤作用下对苯唑西林钠耐受的影响,用基因转录组学分析牛角地黄汤对金黄色葡萄球菌基因转录的变化,用PCR进行验证。【结果】牛角地黄汤与苯唑西林联用能提高金黄色葡萄球菌对苯唑西林的敏感性,牛角地黄汤可抑制耐药相关基因femB、细胞壁合成的相关基因glmUmurG的转录,但对青霉素结合蛋白B(pbpB)及其受体(pbpBR)基因的转录无显著影响。【结论】牛角地黄汤可提高耐甲氧西林金葡菌对苯唑西林的敏感性。

Abstract: 【Objective】 This study was aimed to evaluate the effect of Niujiaodihuang decoction (NDD) on the susceptibility of Staphylococcus aureus (S. aureus) against methicillin. 【Method】 The effect of NDD on the tolerance of S. aureusto oxacillin sodium was detected by microdilution method. The gene transcrip-tional changes of S. aureus were analyzed by gene transcriptional analysis, and the transcriptional changes of the related resistance genes were verified by PCR. 【Result】 The combined effect of NDD and oxacillin improved the inhibitory effect of oxacillin on the proliferation of S. aureus, while inhibiting the transcripti-on level of femB, one of theresistance-related genes, as well as the cell wall synthesis related genes glmU and murG. However it had no significant effect to the transcription of the beta lactamase (pbpB) and its rec-eptor (pbpBR). 【Conclusion】 NDD can increase the sensitivity of methicillin resistant S. aureus to oxacillin sodium. This effect may be related to the reduction of the expression of the gene femB,glmU and murG.
