匙吻鲟表皮葡萄球菌分离鉴定与药敏试验

Isolation, Identification and Drug Susceptibility Test of Staphylococcus epidermidis from Polyodon spathula

摘要: [目的] 对患病匙吻鲟进行致病菌分离鉴定和药敏试验,为匙吻鲟暴发性出血病防治提供一定的参考。[方法] 从患病匙吻鲟分离细菌、形态观察、生理生化试验、16S Sr DNA和PCR引物特异性检测,对致病菌进行分离鉴定,通过K-B法进行药敏试验。[结果] 从发病匙吻鲟病灶组织分离到1株致病菌菌株Pol15001,为γ溶血、革兰氏阳性球菌,生化反应结果、16S rDNA和PCR引物特异检测与表面葡萄球菌相似性均较高,药敏实验结果显示其耐药性较强,对复方新诺明、克林霉素、氨苄西林等7种受试药物耐受,仅对头孢曲松、头孢西叮、氯霉素、头孢噻肟等4种受试药物敏感。[结论] 匙吻鲟为表面葡萄球菌一种新发现的宿主,生产上可通过改善水质及使用头孢曲松、头孢西叮、头孢噻肟等头孢类药物进行防治。

Abstract: [Objective] This study was carried out to isolation, identification and drug susceptibility test of pathogenic bacterium from Polyodon spathula, for providing a reference for prevention and treatment of hemorrhagic disease of Polyodon spathula. [Methods] Bacteria isolation from diseased paddlefish, bacteria morphological observation, physiological and biochemistry reaction by biology automatic microbiological assay system, sequencing and analysis of 16S rDNA, PCR primers specificity testisolation, and identification of pathogenic bacterium, finally the drug susceptibility test were performed with Kirby-Bauer disc diffusion method. [Result] One strain Staphylococcus epidermidis from tissues of diseased and dying Polyodon spathula, named Pol15001, identified based on the bacterial culture, The results showed Staphylococcus epidermidis were resistant to 7 kinds of drugs including selectrin trime-sulf, clindamycin, ampicillin, etc. While sensitive to ceftriaxone, mefoxin, chloroamphenicol and cef otaxime. [Conclusion] The Staphylococcus epidermidis was a new pathogenic bacteria of Polyodon spathula. Improving the aquaculture water quality and taking ceftriaxone, mefoxin, chloroamphenicol and cefo-taxime are effective preventions and controls in aquaculture.
