Genetic Diversity of Brassica napus in Resistance to Sclerotinia sclerotiorum Based on SCOT Markers

摘要: [目的] 探索SCOT标记在油菜遗传多样性研究中的应用。[方法] 以8份具有代表性的甘蓝型油菜品种(系),对130个SCOT引物进行筛选。综合考虑多态性的大小,条带的清晰度,筛选出40个SCOT引物作为核心引物,对43份在田间接种鉴定表现抗菌核病的油菜材料进行遗传多样性分析。[结果] SCOT标记共检测到372个位点,每个引物可以检测到5~12个多态性位点,多态性信息含量值(PIC)介于0.25~0.99,平均值为0.65。供试材料的遗传相似系数集中在0.60~0.88之间。遗传相似度较高,亲缘关系较近。[结论] SCOT标记可以用于油菜遗传多样性的研究。

Abstract: [Objective] The aim of the study was to explore the utilization of start codon targeted polymorphism(SCOT) molecular marker technique in genetic diversity assessment in Brassica napus. [Method] 8representative rape materials were used as templates and 130 SCOT marker primers were screened out. 40 of them showed specific bands and highly polymorphism. The genetic diversity of 43 Sclerotinia sclerotiorum resistant rape materials that was identified in the field were analyzed. [Results] Total 372 alleles were verified using SCOT markers and? about approximately 5-12 alleles were detected by each SCOT marker. Genetic polymorphism information content of each SCOT locus was between 0.25 to 0.99, with a mean value of 0.65. The genetic similarity coefficient ranged from 0.60 to 0.88. Tenetic diversity level of these tea materials was relatively high in this study. [Conclusion] SCOT marker is suitable to explore the genetic diversity of rape germplasm.
