泸州老窖不同窖龄窖泥中己酸菌遗传多样性及系统发育

Genetic Diversity and Phylogeny of Caproic Acid Bacteria in Different Aged Luzhou Laojiao Pit Mud

摘要: [目的] 研究泸州老窖窖泥中己酸菌的遗传多样性和系统发育地位,筛选优良高产己酸菌株,为提高新窖酒品质提供优质菌株资源。[方法] 采用BOXAIR-PCR、16S rDNA PCR-RFLP和代表菌株16S rDNA序列分析技术等对不同窖龄窖泥中乙酸菌的多样性进行分析。[结果] 从不同窖龄窖泥样品中分离获得了43株产己酸细菌;BOXAIR-PCR分析结果表明,在82.2%的相似性水平处,供试菌株形成了8个BOX遗传群;16S rDNA PCR-RFLP将43株供试菌株分为8个遗传群;代表菌株16S rDNA序列系统发育分析表明,供试菌株分布于3个系统发育分支。[结论] 泸州老窖窖泥中己酸菌具有丰富的遗传多样性,这些菌株主要分布于梭菌属(Clostridium)、芽孢杆菌属(Bacillus)和假单胞菌属(Pseudomonas),其中芽孢杆菌属(Bacillus)为优势属。

Abstract: [Objective] The aim of this study was to explore the genetic diversity and phylogeny of cultured caproic acid bacteria in the Luzhou Laojiao pit mud and to screen some effective caproic acid strains. [Method] BOXAIR-PCR, 16S rDNA PCR-RFLP and 16S rDNA sequencing were used to explore the genetic diversity and phylogeny of caproic acid bacteria in different aged Luzhou Laojiao pit mud. [Results] 43 caproic acid strains were isolated from different aged Luzhou Laojiao pit mud;BOXAIR-PCR analysis showed that all strains were 8 BOXAIR-PCR groups at the level of 82.2% similarity. The results of 16S rDNA PCR-RFLP showed that there were eight genotypes among strains. These strains constructed three phylogenic branches. [Conclusion] These caproic acid bacteria belonged to three different genus strains including Bacillus, Clostridium and Pseudomonas and Bacillus was the predominant group. The genetic diversity of caproic acid bacteria was high in the Luzhou Laojiao pit mud.
