山东丹参细胞悬浮培养体系及生长模型的建立

Establishment of Cell Suspension Culture System and Growth Model Salvia miltiorrhiza from Shandong

摘要: [目的] 建立和了解山东丹参疏松愈伤组织和悬浮细胞生长规律。[方法] 以叶片为外植体研究疏松愈伤组织诱导增重以及细胞悬浮培养体系,并对疏松愈伤组织和悬浮细胞进行生长模型的拟合。[结果] 疏松愈伤组织最佳诱导和增殖培养基为MS+2,4-D 0.5mg/L+KT 1.0mg/L+蔗糖30g/L+琼脂6g/L+酸水解酪蛋白1g/L,细胞悬浮培养基为MS+2,4-D 0.5mg/L+KT 1.5mg/L+6-BA 3.0mg/L+NAA 1.0mg/L+蔗糖30g/L+酸水解酪蛋白1g/L。[结论] 山东丹参疏松愈伤组织和悬浮细胞生长曲线为"S"型,Richards方程模型能够很好地描述疏松愈伤组织和悬浮细胞生长曲线变化。

Abstract: [Objective] The aim of the study was to understand the growth characteristics of loose calls and suspension cell of Salvia miltiorrhiza of Shandong. [Method] In this study, leaf was taken as the explants for studying the techniques of loose callus induction and cell suspension culture and growth model of loose callus and suspension cell were fitted by MATLAB 2010B. [Results] The optimal medium for loose callus induction and multiplication was MS+2,4-D 0.5mg/L+KT 1.0mg/L+sucrose 30g/L+agar 6g/L+acid hydrolyzed casein 1g/L. In addition, the optimal medium for cell suspension culture was MS+2,4-D 0.5mg/L+KT 1.5mg/L+6-BA 3.0mg/L+NAA 1.0mg/L+sucrose 30g/L+agar 6g/L+acid hydrolyzed casein 1g/L. [Conclusion] The growth of loose callus and suspension cell exhibited an S-shaped curve. The Richards function model can fit growth dynamics of loose calls and suspension cell of Salvia miltiorrhiza from Shandong.
