一株对鳞翅目高毒力的苏云金芽孢杆菌亚种的分离与鉴定

Isolation and Identification of One Bacillus thuringiensis subsp. Highly Toxic to Lepidoptera

摘要: 从四川温江地区采集土壤,采用醋酸钠-抗生素分离法分离到一株苏云金芽孢杆菌,经形态、生理生化测定,该菌为苏云金芽孢杆菌莫里逊亚种(Bacillus thuringiensis subsp. morrisoni)。生物测定结果表明,该菌对菜青虫表现出较高的毒力,在饲喂浸含伴孢晶体的叶片72 h后校正死亡率达到96.3%。根据Genbank中的序列设计cry基因型鉴定引物,对此菌种进行cry基因型检测,结果表明,该菌含有对鳞翅目高毒力的cry1Ac、cry1Ab和cry1C基因,还含有对植物寄生线虫高毒力的cry5基因。

Abstract: A strain of Bacillus thurigiensis was isolated from soils in Wenjiang in Sichuan by Sodium acetate-antibiotic method. According to identification, this strain belongs to Bacillus thuringiensis subsp. morrisoni. Bioassay shows that this strain has high insecticidal activity against Pieris rapae L., and the corrected mortality is 96.3% 72 h after feeding with leaves immersed in cultures containing crystalline parasporal body. Primers are then designed to identify the genotype of cry genes. The results indicate that this strain contained cry1Ac, cry1Ab, cry1C highly toxic to Lepidoptera and cry5 genes highly toxic to Pratylenchus spp..
