Determination of GA3,IAA,ABA and ZT in Dormant Buds of Allium ovalifolium by HPLC

摘要: 以卵叶韭休眠芽为试材,改进了提取纯化赤霉素(GA3)、生长素(IAA)、脱落酸(ABA)和玉米素(ZT)4种内源激素的处理方法和色谱条件。采用Hypersil ODS C18柱(150 mm×4.6 mm,5μm),甲醇-0.6%冰乙酸进行梯度洗脱,紫外检测波长254 nm。实际分析样品表明,本法分离度好,准确度高,适合大批量测定。

Abstract: An improved method of extraction,purification and determination to four internal hormones(GA3,IAA,ABA and ZT) in dormant buds of Allium ovalifolium with Reversed-phase High Performance Liquid Chromatography has been developed.Chromatographic conditions are:Hypersil ODS C18 reverse phase column(150 mm×4.6 mm,5 μm),methanol-0.6% acetic acid mobile phase,gradient elution,UV detector set at 254 nm.The result shows that this methods is a accurate and high-separation-efficiency technique,suitable to determine large quantities of samples.
