长寿花离体繁殖技术研究

Study on in Vitro Propagation of Kalanchoe blossfeldiana

摘要: 用长寿花的带芽茎段为外植体,进行了外植体的建立、增殖、生根及移栽研究。结果表明:芽启动培养基以MS+6-BA0.5mg/L+NAA0.1mg/L为宜,启动率达100.0%。增殖培养中,6-BA与NAA、GA配合使用增殖效果好,外植体培养在MS+6-BA2.0mg/L+NAA0.2mg/L+GA30.1mg/L上可增殖9.3倍。采用MS+6-BA1.0mg/L+GA30.1mg/L+AC2.0g/L的半固体培养基,可使长寿花增殖与生根同步进行,每个外植体平均可产生5.5个芽,生根率达100.0%。试管苗移栽入土成活率达100.0%。

Abstract: Stems with buds of Kalanchoe blossfeldiana were used as explants. The establishment of explants, proliferation and rooting of shoots and transplantation of plantlets were studied. The results show that the optimum initiation medium is MS+6-BA 0.5 mg/L+NAA 0.1 mg/L, with 100.0% initiation rate obtained. Shoots multiply more effectively on the media of combinations of 6-BA, NAA and GA3. The shoots cultured on the medium of MS+6-BA 2.0 mg/L+NAA 0.2 mg/L+GA3 0.1 mg/L result in 9.3 times of proliferation. The semisolid medium of MS with 6-BA 2.0 mg/L, GA3 0.1 mg/L and AC 2.0 g/L is more effective than solid medium for the multiplication and rooting for shoots, each explant produces up 5.5 seedlings and all the explants root successfully. The plantlets are transplanted into nutrient soil with 100.0% survival ratio.
